摘 要:目的:观察柔肝通络汤对脑缺血大鼠内皮祖细胞(Endothelial Progenitor Cells,EPCs)的影响,探讨其改善脑缺血神经功能缺损的可能机制。方法:采用线栓法制备大脑中动脉闭塞的脑缺血大鼠模型,随机分为0.9%氯化钠注射液组、辛伐他汀组、柔肝通络汤组,每组各10只;另取10只设为假手术组。各组分别于造模成功后第1天开始灌胃给药,连续7d。采用Longa神经功能评分评价造模术后3h及给药前后各组大鼠神经功能缺损情况,于造模前及给药前后使用CD_(34)、CD_(133)双抗体标记结合流式细胞仪检测各组大鼠外周血EPCs数量。结果:各治疗组给药前Longa评分均较造模术后3h降低;给药后Longa评分柔肝通络汤组、辛伐他汀组均低于0.9%氯化钠注射液组,且柔肝通络汤组低于辛伐他汀组,差异均有统计学意义(P<0.01);急性脑缺血发生后,大鼠外周血CD_(34)+CD_(133)+EPCs数量先下降再上升,柔肝通络汤与辛伐他汀可使EPCs数量增加,且柔肝通络汤作用优于辛伐他汀。结论:柔肝通络汤可能通过增加EPCs数量及功能来促进大鼠脑缺血后神经功能的恢复。Objective:To investigate the effect of Rougan Tongluo decoction on endothelial progenitor cells(EPCs) in rats with cerebral ischemia and its possible mechanism in improving neurological defects induced by cerebral ischemia.Methods:The suture method was used to establish a rat model of cerebral ischemia with middle cerebral artery occlusion.The rats were randomly divided into 0.9% sodium chloride injection group,simvastatin group,and Rougan Tongluo decoction group,with 10 rats in each group,and another 10 rats were selected as sham-operation group.The drugs were given by gavage from the first day after the model was established successfully for 7 consecutive days.The Longaneurological score was used to evaluate neurological defects at 3 hours after modeling and before and after administration,and CD_(34)、CD_(133) double-antibody labeling combined with flow cytometry was used to measure the number of peripheral EPCs before modeling and before and after administration.Results:All the treatment groups had a significant reduction in the Longaneurological score before administration.After administration,the Rougan Tongluo decoction group and the simvastatin group had a significantly lower score than the 0.9% sodium chloride injection group,and the Rougan Tongluo decoction group had a significantly lower score than the simvastatin group(P0.01).After the onset of acute cerebral ischemia,the number of peripheral CD_(34)+CD_(133)+EPCs decreased at first and then increased;bothRougan Tongluo decoction group and simvastatin group had an increase in the number of EPCs,and the Rougan Tongluo decoction group had a significantly greater increase than thesimvastatin group.Conclusion:Rougan Tongluo decoction can promote the recovery of neurological function after cerebral ischemia in rats by increasing the number of EPCs.
