摘 要:比较2型猪链球菌(S.suis 2)ΔRevS突变株和05ZY/H33野生株生物学特性及致病性的差异.首先,通过吸光值测定和革兰染色比较△RevS突变株和05ZY/H33野生株的生长速度和菌体形态.分别对Balb/c及ICR(CD1)小鼠腹腔注射109 CFU·mL^(-1)的05ZY/H33菌液,评估两种品系小鼠对S.suis 2的易感性.分别对Balb/c小鼠腹腔注射2.5×10~9,5.0×10~8,1.0×108,2.0×10~7,4.0×10~6 CFU·mL^(-1)的05ZY/H33菌液,采用Reed法计算半数致死量.对Balb/c小鼠腹腔注射5.0×108 CFU·mL^(-1)的ΔRevS突变株和05ZY/H33野生株菌液,观察野生株和突变株对小鼠的影响.对仔猪耳缘静脉注射10~9 CFU·mL^(-1)的05ZY/H33野生株和ΔRevS突变株菌液,观察野生株和突变株对仔猪的影响.体外生物学特征分析表明,与野生株相比,突变株的生长速率和形态染色并未发现显著变化;Balb/c小鼠对S.suis 2 05ZY/H33的敏感性高于ICR(CD1)小鼠,05ZY/H33对Balb/c小鼠的LD50为4.2×10~7 CFU·mL^(-1).Balb/c小鼠攻毒试验结果显示,ΔRevS对小鼠的致病性明显减弱.仔猪攻毒试验结果显示,野生株和突变株都引发仔猪全部死亡,但ΔRevS引发仔猪死亡时间较晚.△RevS突变株保持了菌株的基本生物学特征,但对小鼠和仔猪两种动物模型的致病作用差异较大.Compare the biological characteristics and pathogenicity difference between△RevS mutant and 05ZY/H332 wild strains of S. suis 2. First of all, compare the growth rate and bacterial morphology between △RevS mutant and 05ZY/H332 wild strains by absorbance measurement and gram stain. To evaluate the susceptibility of different strains, Balb/c and ICR (CD1) mice were injected intraperitoneally with 109 CFU · mL 05ZY/H33 solution. Balb/c mice were injected intraperitoneally with 2.5× 10^9 , 5.0× 10^8 , 1.0 ×10^8 , 2.0 × 10^7 , 4.0× 10^6 CFU ·mL 1 05ZY/H33 solution, then Reed method was used to calculate LDs0. To study the effects of wild and mutant strains on mice, Balb/c mice were injectedintraperitoneally with 5.0 × 10^8 CFU ·mL 1 2~RevS solution and 05ZY/H33 solution. To study the effects of wild and mutant strains on piglet, piglets were injected intraperitoneally with 109 CFU · mL-1 05ZY/H33 solution and △RevS solution. Compared with wild strains, the growth rate and morphology of the mutant were not significantly changed. The sensitivity of Balb/c mice to S.suis 2 05ZY/H33 was higher than ICR(CD1) mice, LD50 of 05ZY/H33 to Balb/c mice was 4.2× 10^7CFU ·mL-1. The poison test results on Balb/c mice showed the pathogenicity of △RevS mutant was obviously decreased. The poison test results on piglets showed that both wild strains and mutant strains caused the death of piglets, but the time of death of △RevS mutant was later. The △RevS mutant strains maintained the basic biological characteristics of the strain, but the pathogenic effect of the two animal models of mice and piglets was significantly different.
