摘  要:目的:探讨覆盆子水煎剂对H_2O_2诱导的小鼠脑微血管内皮细胞(b End.3细胞)氧化应激损伤的影响。方法:用不同浓度的H_2O_2处理b End.3细胞6 h,构建氧化应激模型;MTT法检测不同浓度的覆盆子水煎剂对b End.3细胞活性的影响;采用Western blot法检测相关凋亡蛋白Bax、Bcl-2、Cytochrome(Cyt-c)、Caspase 3、Caspase 8、Caspase 12蛋白的表达情况。结果:MTT检测发现覆盆子水煎剂可提高H_2O_2干预下的b End.3细胞活性,并且该保护作用呈现浓度依赖性(P<0.05)。Western blot检测结果显示覆盆子水煎剂可抑制Cyt-c高表达,上调Bcl-2/Bax比值,同时下调Cleaved-caspase 3/pro-caspase 3、Cleaved-caspase 8/pro-caspase 8、Cleaved-caspase 12/pro-caspase 12的比值(P<0.05)。结论:实验剂量的覆盆子水煎剂对H_2O_2介导的b End.3细胞损伤具有保护作用,主要与调节相关凋亡蛋白Bcl-2、Bax、Cyt-c的表达及Caspase 3、8、12活化有关。Objective: To investigate the effect of Raspberry decoction on hydrogen peroxide-treated mouse brain microvascular endothelal cells (bEnd.3) induced injury. Methods: bEnd.3 cells were treated with H2O2 of different concentrations for 6 h to establish the hydrogen peroxide-treated bEnd.3 model. MTT (Tetramethyl tria- zole salt) assay was used for checking the cell viability. The Western blot method was used for detecting Bax, Bcl-2, Cytochrome (Cyt-c), Cleaved-caspase 3, Cleaved-caspase 8 and caspase 12 protein expressions. Results: MTT revealed the cells survival rate was increased with concentration-dependence in vitro (P〈0.05). Western blotting revealed that the expression of Bax, Cyt-c, Cleaved-caspase 3/pro-Caspase3, Cleaved-caspase8/pro Cas- pase-8, Cleaved-caspase12/pro-Caspasel2 was decreased, and the expression of Bcl-2 increased by different concentration of Raspberry decoction(P〈0.05). Conclusion: The Raspberry aqua decoction was able to protect bEnd.3 cells from apoptosis induced by H^O2, mainly through influencing proteins expression of Bcl-2, Bax, Cyt- c, and activation of Caspase3, 8, 12.
