摘 要:目的:观察黄连解毒汤对脑缺血大鼠模型脑组织中NF-Kbp65动态变化的影响,探讨黄连解毒汤对脑缺血的作用机制。方法:清洁级SD大鼠240只,按随机数字表法随机分为假手术组、模型组、低剂量黄连解毒汤组、中剂量黄连解毒汤组和高剂量黄连解毒汤组,每组48只。造模成功后即予不同剂量的黄连解毒汤干预,分别在1 h、12 h、24 h、72 h、6 d、15 d时间点每次取8只大鼠断头去脑,分离脑组织,4只用于免疫组化染色、4只用于RT-PCR检测NF-Kbp65含量变化。结果:模型大鼠脑组织中NF-Kbp65含量明显增加,不同剂量黄连解毒汤均能显著降低脑组织中NF-Kbp65的含量(P<0.05),以高剂量黄连解毒汤组降低最明显。结论:黄连解毒汤可降低脑缺血模型大鼠脑组织中NF-Kbp65含量,可能是该方治疗脑缺血的作用机制之一。Objective: To observe the influence of Huanglian Jiedu decoction on NF-Kbp65 dynamic change in cerebral ischemia model rats, in order to explore the mechanism of Huanglian Jiedu decoction on cerebral ischemia. Methods: A total of 240 clean level SD rats were randomly divided into control group, model group, low dose of Huanglian Jiedu decoction group, medium dose of Huanglian Jiedu decoction group and high dose of Huanglian Jiedu decoction group, 48 rats in each group. Different dosage of Huanglian Jiedu decoction was given. In 1 h, 12 h, 24 h, 72 h, 6 d, 15 d, 8 rats were killed to separate the brain tissues respectively, and 4 were used for immunohistochemical staining, and the other 4 rats were used for RT- PCR to detect NF-Kbp65 content changes. Results: The NF-Kbp65 levels increased in model group significantly. Different doses of Huanglian Diedu decoction can decrease the NF-Kbp 65 levels (P〈0.05), and the high dose of Huanglian Jiedu decoction group showed best effect. Conclusion: Huanglian Jiedu decoction can reduce NF-Kbp65 content in cerebral ischemia model rat; it may be one of the mechanisms for the treatment of cerebral ischemia.
