摘  要:目的:建立测定肾八味胶囊中黄酮类化合物含量的方法。方法:采用HPLC法,样品用双蒸馏水分散后,超声提取、离心,用Inertsil ODS-3色谱柱(250 mm×4.6 mm,5μm)分离化合物,流动相为乙腈与水,梯度洗脱,流量为1 mL·min-1,柱温为室温,检测波长为215 nm,外标法定量。结果:肾八味胶囊用加样回收法与对照品比较,检出黄酮类化合物5个,芦丁、野黄芩苷、金丝桃苷、黄芩素、汉黄芩素;样品中多数峰可以达到良好分离;5个测定化合物在各自的浓度范围内均有良好的线性(r>0.999),方法回收率分别为芦丁(101.31±2.14)%、野黄芩苷(102.19±1.5)%、金丝桃苷(100.48±2.97)%、黄芩素(100.47±2.78)%、汉黄芩素(100.75±2.54)%。结论:本方法可用于肾八味胶囊中黄酮类化合物含量测定。Objective: To develop a HPLC-UV method for determination the flavonoids content of Shenbawei capsule. Methods: Samples were dissolved in water and were used ultrasonic extraction, centrifugal. The separation was performed on Inertsil ODS-3 column (250 mm×4.6mm,5μm) column with mobile phase consisted of Acetonitrile and water (gradient elution), flow rate was 1.0 mL·min-1, at room temperature and detected with UV detector at 215 nm. External standard method was quantified. Results: As compared with standard sample, Rutin, Scutellarin, Hyperoside, Baicalein, Wogonin were checked out. Most peaks were well separated. The five compoundshowed a good linearity within the range of the respective concentrations (r〉0.999). The method recoveries are Rutin (101.31±2.14)%, Scutellarin (102.19±1.5)%, Hyperoside (100.48±2.97)%, Baicalein (100.47±2.78)%, Wogonin (100.75±2.54)% respectively. Conclusion: The method can be used to determine the flavonoids content of Shenbawei capsule.
