摘  要:目的:观察滋水清肝饮对内分泌治疗后的乳腺癌移植小鼠脑内单胺类神经递质的影响,探讨其改善绝经期综合征的机制。方法:在40只BALB/c雌性小鼠右侧腹部倒数第二对乳腺脂肪垫下分别接种5×105个4T1细胞,建立乳腺癌移植小鼠模型,再通过去卵巢手术建立绝经后同种乳腺癌移植小鼠模型,将小鼠随机分4组,分别为:实验组1(低剂量组):滋水清肝饮11.28g·kg^(-1)+来曲唑0.379mg·kg^(-1)1;实验组2(高剂量组):滋水清肝饮45.1g·kg^(-1)+来曲唑0.379mg·kg^(-1);空白对照组:等量蒸馏水;阳性对照组:来曲唑0.379mg·kg^(-1)。各组分别灌胃给药4周后麻醉处死,酶联免疫吸附测定(ELISA)法分别测定各组小鼠腹主动脉血中雌二醇(E2),促卵泡刺激素(FSH),促黄体生成素(LH)含量及脑内海马区5-HT、NE含量;蛋白质免疫印迹(Western Blot)检测5-HT1AR、5-HT2AR蛋白表达量。结果:ELISA结果显示,与阳性对照组相比,高剂量组滋水清肝饮能显著能降低小鼠体内E2,FSH,LH含量(P<0.05);并能升高小鼠脑内5-HT、NE水平(P<0.05);Western Blot结果显示,与空白对照组及阳性对照组比较,高、低两种剂量滋水清肝饮均能上调脑内5-HT1AR蛋白水平,下调5-HT2AR蛋白水平(P<0.05),且高剂量组对5-HT1AR及5-HT2AR的影响更加明显(P<0.05)。结论:滋水清肝饮可能通过调节单胺类神经递质缓解内分泌治疗后的绝经期综合症。Objective: To observe the effect of Zishui Qingan Decoction on monoamine in the brain after endocrine therapy for breast cancer transplanted mice, and to explore its mechanism to improve menopausal syndrome. Methods: 5- 1054Tlcells were inoculated in the second and lower breast fat cubes of the right abdomen of 40BALB/c female mice, and the model of breast cancer transplant was established. The postmenopausal breast cancer transplant was established by ovariectomy. The mice were randomly divided into 4groups: experiment group 1 (low dose group) : Zishui Qingan Decoction 11.28g·kg-1 + letrozole 0. 379mg ·kg-1 ; experimental group 2 (high dose group) : Zishui Qingan Decoction 45.1g·kg-1 +letrozole 0. 379mg·kg-1 ; blank controlled group: equal volume of distilled water; positive controlled group: letrozole 0. 379mg·kg-1 , after estrous administration for 4weeks, the levels of estradiol (E2), follicle stimulating hormone (FSH) and luteinizing hormone (LH) were measured by enzyme-linked immunosorbent assay (Elisa) ; The contents of 5-HT1AR and 5-HT2AR protein were detected by Western blotting; The expression of 5-HT1AR and 5-HT2AR protein were detected by Western blotting. Results: Elisa method showed that the dose of E2, FSH and LH in mice was significantly lower than that in the positive controlled group (P〈0. 05), and the levels of 5-HT and NE (P〈0. 05). Compared with the model group and the positive controlled group, the expression of 5-HT1AR increased and the expression of 5-HT2AR was decreased (P〈0. 05), and the expression of 5-HT1AR in the brain was higher than that in the blank controlled group and positive controUed group, and the effect of high dose group was' more obvious (P〈0. 05). Conclusion: Zishui Qinggan Decoction may relieve endocrine therapy-related menopausal syndrome by modulating monoamine neurotransmitters.
