摘 要:为探究褪黑素对大豆幼苗耐盐性的调节作用,明确褪黑素提高大豆耐盐能力的作用途径。以大豆品种天隆2号为材料,通过水培试验,分析了盐胁迫下施加外源褪黑素(200μmol/L)对大豆幼苗生长、活性氧代谢及渗透调节物质的影响。结果表明,在盐胁迫下,大豆的生长受到了抑制,显著增加了过氧化氢(H2O2)与丙二醛(MDA)含量、可溶性蛋白(WSP)含量、超氧化物酶(SOD)和过氧化物酶(POD)活性。在盐胁迫下施加外源褪黑素可缓解大豆幼苗的盐毒害症状,增加其生物量,与盐处理相比,显著地提高了抗氧化酶(SOD和POD)活性,减少了H2O2与MDA的积累,同时渗透调节物质WSP含量也显著上升。此外,正常水培条件下,褪黑素对大豆幼苗的生长也有促进作用。以上结果说明,在盐胁迫下,施加外源褪黑素可以通过增加大豆幼苗抗氧化酶活性来清除过多的H2O2,减轻膜脂过氧化反应,并通过调节渗透物质缓解盐胁迫诱导的渗透胁迫,促进大豆幼苗生物量积累,从而提高大豆幼苗耐盐能力。Melatonin is a naturally occurring compound in plants,and this study is to reveal the effects of melatonin on soybean seedlings under salt stress. The experimental material,Tianlong 2 was planted in nutrient solution,and melatonin( 200μmol/L) was applied. The traits of seedlings growth,active oxygen metabolism and osmotic adjustment substance were investigated. The results showed that salt stress inhibited the seedling growth,resulted in H_2O_2 and MDA accumulation,increased WSP content and enhanced the activity of SOD and POD significantly. Under salt stress condition,exogenous melatonin could alleviate the symptoms of salt toxicity and increase the biomass of soybean seedlings. The activity of antioxidant enzymes( SOD and POD) was improved significantly by exogenous melatonin,and the content of H2O2 and MDA was decreased. The results also showed that solute accumulation under stress was significantly enhanced with exogenous melatonin by increasing the content of solubleproteins. Moreover,melatonin also could promote the growth of soybean seedlings under normal conditions. This study indicated that exogenous melatonin could improve the H_2O_2-scavenging capacity of soybean seedlings by enhancing the activity of antioxidant enzymes and could also alleviate osmotic stress by promoting the accumulation of osmoregulatory substances. Ultimately,exogenous melatonin improved the biomass of soybean seedlings grown under salt stress,thereby effectively alleviating salt stress in soybean seedlings.
