白花蛇舌草水提物对秀丽隐杆线虫急性毒性的研究

摘　　要：目的:研究白花蛇舌草不同浓度水提取物对野生型秀丽隐杆线虫的毒性作用。方法:用水对白花蛇舌草进行回流提取,提取物挥干后用DMSO溶解配制成不同浓度的溶液,对照组为培养液,用不同浓度的白花蛇舌草水提取物对野生型秀丽线虫幼虫进行暴露处理,每隔一定时间观察药物暴露后的秀丽隐线虫的存活率,体长,运动行为,开始怀卵时间等生理指标,与对照组比较,分析不同浓度药物对秀丽隐杆线虫的毒性影响。结果:与对照组比较,48 h处理后,白花蛇舌草水提取物浓度1.00 mg·mL^(-1)以上有明显致死作用,实验组线虫体长缩短,开始怀卵时间推迟,头部运动频率和身体弯曲程度均受到影响。结论:白花蛇舌草水提取物对秀丽线虫的生殖及发育有毒性作用,浓度越高,毒性作用越明显。Objective： To study the toxicity of different concentration of Oldenlandia diffusa aqueous extracts to wild Caenorhabditis elegans（C.Elegans）.Methods： Wild C.Elegans were exposed into different concentration extracts of Oldenlandia diffusa which refluxed by water and were dissolved in DMSO.The control group was disposed into nutrient solution.The survival rate,body length,motor behavior,and the time of ovulation were observed at regular intervals.The data of the two groups were compared.The toxicity effects of different concentrations of drugs on the o C.elegans were analyzed.Results： Compared with the control group,after 48 hours treatment,there was a significant lethal effect of Oldenlandia diffusa when the concentration of its aqueous extract was above 1.00 mg· mL-1,which led to the results that the length of nematode was shortened,the beginning time of pregnancy was delayed,and the head movement frequency and the degree of physical bending were affected.Conclusion： The aqueous extracts of Oldenlandia diffusa has toxic effects on the reproduction and development of the C.elegans.The higher concentration is,the more obvious the toxic effect.
