应用RACE法克隆雷竹笋PAL基因及序列分析

摘  要:应用RACE法,克隆获得了雷竹笋PAL基因的全长序列,并对其进行了生物信息学分析。结果表明:雷竹笋PAL基因的开放阅读框长度为2103 bp,共编码701个氨基酸,其中具有1个保守的活性位点Ala-Ser-Gly(A-S-G);在PAL蛋白的二级结构中含有大量的α螺旋和β折叠,其三级结构模型为一个带柄的圆锥形。构建的PAL基因核苷酸序列进化树结果显示雷竹与毛竹、绿竹的亲缘关系最近,与小麦、大麦、水稻、佛肚竹的亲缘关系较近,与玉米、高粱的亲缘关系较远。RACE method was applied to clone and obtain the full-length sequence of PAL gene from bamboo shoot(Phyllostachys praecox),and its bioinformatics analysis was carried out.The results indicated that:the open reading frame of the cloned PAL gene from bamboo shoot was 2103 bp in length,it encoded 701 amino acids,and had a conservative activity site Ala-SerGly(A-S-G); there were a large number of α-helix and β-sheet in the secondary structure of PAL protein,and the tertiary structure model of PAL protein was a tapered cone with a handle.The phylogenetic tree analysis of nucleotide sequences in PAL gene showed that P.praecox had the closest affinity relationship with Phyllostachys edulis and Bambusa oldhamii,had a closer affinity relationship with Triticum aestivum,Hordeum vulgare,Oryza sativa and Bambusa ventricosa,and had a farther affinity relationship with Zea mays and Sorghum bicolor.
