摘 要:目的观察银杏叶提取物、对重复正加速度(+G_z)暴露高脂血症大鼠主动脉凝集素样氧化低密度脂蛋白受体-1(oxidized low-density lipoprotein receptor-1,LOX-1)及核因子-κB(NF-κB)表达的影响。方法雄性SD大鼠50只随机分为5组,对照组、+Gz作用高脂模型组、银杏叶提取物低剂量干预组、银杏叶提取物中剂量干预组和银杏叶提取物高剂量干预组,每组10只。对照组给予普通饲料,模型组和药物干预组大鼠均给予高脂饲料喂养合并重复+Gz暴露处理,药物干预组大鼠每日灌胃给予各自剂量银杏叶提取物1次。实验持续8周后,检测各组大鼠血脂水平、氧化应激指标、炎症反应指标以及胸主动脉LOX-1、NF-κB表达。结果银杏叶提取物能降低重复+Gz暴露合并高脂饮食大鼠氧化型低密度脂蛋白水平,下调主动脉LOX-1表达,抑制NF-κB活化,减少促炎症因子的产生。结论银杏叶提取物可能通过下调主动脉LOX-1表达,抑制NF-κB活化,减轻炎症反应,进而减轻重复+Gz暴露合并高脂饮食导致的主动脉炎症损伤。Objective To investigate the effect o f Ginkgo biloba ex tract (GBE) on the expression o f LOX-1 and NF-κB in aortas of high-fat diet-induced hyperlipidemic rats exposed to repeated positive acceleration. Methods Fifty male Sprague-Dawley rats were randomly allocated to five groups, including a control group, a high lipid with repeated positive acceleration stress group, a low dosage group of GBE, a middle dosage group of GBE, and a high dosage group of GBE. The rats of control group were fed with common feed and those of other groups were administered with high lipid feed and exposed to repeated positive acceleration. After eight weeks of intragastric administration with GBE, the serum was separated, and thoracic aortas were kept. The following indicators were observed lipid metabolism indicators, oxidative stress indicators and inflammation reaction indicators, and the protein expressions of LOX-1 and NF-κB. Results GBE red u c ed ox-LDL, d own - reg u la ted LOX-1 ex p res sio n , in h ib i te d N F -κB a c t iv a tio n and decreased the generation of proinflammatory factors. Conclusion GBE can p ro te c t a g a in s t th e in f lam m a t io n of aortas of high-fat diet-induced hyperlipidemic rats exposed to repeated positive acceleration by down-regulating LOX-1 expression and inhibiting NF -κ B a c tiv a tio n .
