摘 要:目的探讨几种炎症因子在choke区血管新生过程中的作用。方法建立小鼠跨区供血耳瓣模型,切取小鼠耳瓣,HE染色后,观察血管管径变化。运用免疫组织化学技术,分别在造模术后当日(0d)、1d、3d、5d、7d检测细胞间黏附分子-1(ICAM-1)、单核细胞趋化蛋白-1(MCP-1)、巨噬细胞标记物(CD68)的表达情况。结果对小鼠耳瓣血管管径观察显示,术后3d可见新生血管,7 d后血管扩张明显。在术后3d,两个choke区中CD68标记的巨噬细胞聚集量最大,MCP-1和ICAM-1在术后早期炎症反应期间出现高表达。choke区血管扩张期间,组织中的三种炎症因子表达明显增多。结论ICAM-1、CD68、MCP-1三种炎症因子在血管重塑中起重要作用,适当的炎症反应对皮瓣存活起到积极作用。Objective To explore the role of inflammation in angiogenesis in choke area. Methods The ear flap model of mice was established and the changes of blood vessel diameter was observed by HE staining. The expressions of intercellular cell adhesion molecule-1 (ICAM-1), monocyte chemoattractant protein-1 (MCP-1) and cluster of differentiation 68 (CD68) were detected by immunohistochemistry on the day after modeling(0 d), 1d, 3 d, 5 d and 7d, respectively. Result By observing the vascular diameter of the ear flap in mice, it was found that the vessels were visible at 3 d after operation and the vascular dilatation was obvious at 7 d. CD68-1abeled macrophages had the largest accumulation in two choke area at 3 d. High expression of MCP-1 and ICAM-1 was observed in the early stage of postoperative inflammatory response. The expression of the three inflammatory factors in the tissue was increased significantly during the vascular dilation. Conclusion The three inflammatory factors (ICAM-1, CD68 and MCP-1) play an important role in vascular remodeling. Proper inflammatory response plays a positive role in flap survival.
