摘  要:文章分别从不同部位、不同的消毒处理方法、不同的切段方法、以及不同的切段长度4个方面对异枝江蓠(Gracilariabailinae)进行了组织切段培养的研究。研究结果表明,在不同部位的切段实验中,发现主干部位出芽数量显著多于分枝中部与茎尖的出芽数量,为最适合异枝江蓠切段培养的取材部位。在不同消毒方法的实验中,综合成活率与杂藻污染率来看,先经0.5g/L青霉素钠+0.5g/L硫酸链霉素+1mg/L GeO2处理16h,然后用消毒海水培养48h的预处理,再用70%的酒精处理5s或1.5%KI处理10min消毒效果较好。在不同切段方法的实验中,斜切与横切的出芽数差异不显著。在不同切段长度的实验中,长度为2mm的切段芽密度最大,为最经济的切段长度。大多数切段的下切面均未发现有新芽长出,表现出明显的极性。Different measures of tissue culture of Gracilariabailinae were evaluated in the present stud- y. Four various aspects were taken into consideration including positions of G. bailinae, treatments of sterilization,fragment length and cutting methods. The results showed that the number of budding of trunks was significantly higher than those of middlebranchs and stem tips, indicating that trunks is the best position for tissue culture of G. bailinae. The G. bailinae that was steeped with 0.5g/L penicillin sodium,0.5g/L streptomycinsulphate and lmg/L GeO2 for 16h and then was immersed into the steri- lized seawater about 48h, treated with 70% ethyl alcohol for 5s or 1.5%KI treated for 10min,had high- er survival rates and lower contamination rates of wild seaweed than other treatments. There is no significant difference in budding numbers by using different cutting methods. Among different fragment lengths, 2mm fragment is the most economical one that has the biggest bud density. In most of the fragments, budding couldnotn't be found in the lower transection, which showed a significant polarity.
