摘  要:目的观察YKL-40在烟雾诱导的慢性阻塞性肺病(Chronic obstructive pulmonary disease,COPD)大鼠肺组织中的动态表达。方法选取SPF级SD大鼠30只随机分为观察组与对照组,每组15只。对照组呼吸清洁空气,观察组被动吸入香烟烟雾。各组大鼠分别于实验的第4周、8周、12周称重,麻醉处死。通过HE染色判断大鼠模型肺泡炎及气道重塑,采用RT-PCR、Western blot检测YKL-40在大鼠肺组织中的表达。大鼠体重变化比较采用t检验;观察组与对照组组间及各时间点比较采用重复测量方差分析。基因表达与吸烟的相关性采用Pearson相关性分析,P<0.01为差异有统计学意义。结果大鼠被动吸烟3个月成功建立COPD动物模型。在转录水平,随着大鼠吸烟时间的延长,观察组YKL-40 m RNA相对表达量增加:1个月、2个月、3个月观察组YKL-40m RNA与对照组比较差异有统计学意义(均P<0.01)。观察组YKL-40的蛋白表达增强,与对照组比较差异有统计学意义;观察组之YKL-40 m RNA及蛋白表达各时间段比较,差异有统计学意义(均P<0.01)。结论 YKL-40在COPD大鼠肺组织中表达增强,可以作为COPD发生发展的一个生物学标志物。Objective To observe the expression of YKL-40 in the lungs of rats with chronic obstructive pulmonary disease.Methods A total of 30 SPF grade SD rats were randomly divided into observation group and control group, 15 rats in each group. The control group received clean air, and the observation group received passive inhalation of cigarette smoke. Rats in each group were weighed at 4th, 8th and 12th weeks respectively, and they were sacrificed by anesthesia. The alveolar inflammation and airway remodeling were determined by HE staining, and the expression of YKL-40 in lung tissue of rats was detected by RT-PCR and Western blot. T test was used to compare the changes of body weight in rats. The repeated measures variance analysis was used between the observation group and the control group at each time point. P〈0.01 was considered statistically significant.Results COPD animal model was successfully established three months after passive smoking. YKL-40 mRNA relative expression of the observation group increased with the prolongation of smoking time. Compared with the control group, expression of YKL-40mRNA was enhanced in the observation group after 1 months, 2 months and 3 months of passive smoking (all P〈0.01). The expression of YKL-40 protein in the observation group was higher than that in the control group. The expressions of YKL-40 protein at different time points in observation group were statistically significant(all P〈0.01).Conclusion The expression of YKL-40 in lung tissue of COPD rats is enhanced, which can be used as a biological marker for the occurrence and development of COPD.
