包埋脱水法常温和低温保存裙带菜配子体的研究

摘  要:采用包埋脱水法在20℃、4℃和-20℃条件下对裙带菜配子体进行保存,研究了保存温度、胶球含水量以及光照等因素对裙带菜配子体存活率的影响。含水量为50%时配子体在20℃/暗和4℃/暗获得了较好的存活率,保存360 d后存活率分别为22.4%和56.3%。保存360 d的裙带菜配子体可以发育成幼孢子体,且与对照相比无明显差异。结果表明,该方法适合对裙带菜配子体进行中长期保存,该方法操作简单,对裙带菜的保种及育苗有重要意义。Gametophytes of Undaria pinnatifida were preserved by encapsulation-dehydration at 20 ℃,4 ℃ and-20 ℃ to determine the optimal temperature. The results showed that the high viability were obtained under beads water content of 50% at 20 ℃/dark and 4 ℃/dark. After 360 days,the viability could reach about 22. 4% at 20 ℃/dark,56. 3% at 4 ℃/dark. After storage for 360 days,the gametophytes could produce normal sporophytes in the conditions suitable for development. The results indicated that encapsulation-dehydration method is suitable for preservation of the gametophytes,since it is simple in operation which would have important significance for the conservation and breeding of Undaria pinnatifida.
