摘 要:目的 :利用CatWalk动物步态采集分析系统,考察氧化苦参碱(Oxymatrine,OMT)对神经病理性疼痛小鼠模型步态行为的改善及纠正作用。方法 :按随机数字表方法将60只小鼠完全随机分为假手术(Sham)组、模型(Model)组和氧化苦参碱(OMT)组,每组20只。采用部分结扎左侧后肢坐骨神经的方法建立部分坐骨神经结扎(PSNL)神经病理性疼痛模型。各组小鼠经训练后分别进行实时步态行为录像,进行数据采集分析。结果 :与Sham组比较,Model组中,坐骨神经功能指数(SFI)、手术侧后足足印长度(Print Length)、足印宽度(Print Width)、足印面积(Print Area)、最大接触时间的足印面积(Max Contact Area)、站立时间(Stand)、单足接触时间(Single Stance)、最大接触时间的最大强度(Max Contact Max Intensity)及完整足印的平均强度(Mean Intensity),9项指标显著下调(P〈0.01);OMT使Model组中上述9项指标显著上调(P〈0.05)。与Sham组比较,Model组中,手术对侧前足足印长度、足印宽度、足印面积、最大接触时间的足印面积4项指标显著上调(P〈0.01);OMT使Model组中上述4项指标显著下调(P〈0.05);而手术侧后足的身体协调性相关3项指标:步周长(Stride Length)、摆动时间(Swing)、身体速度(Body Speed)及正常步序比指标(Regularity Index)未见任何变化。结论 :OMT对神经病理性疼痛模型小鼠的步态行为有明显改善及纠正作用。Objective :To investigate the improvement and adjustment of oxymatrine(OMT)on the gait behavior of mice with neuropathic pain by CatWalk analysis system. Methods :Sixty mice were randomly divided into sham-operation group,neuropathic pain model group,and OMT group according to random digit table and 20 mice in each group. Neuropathic pain model was established through partial sciatic nerve ligation(PSNL)on the left side of mice. Real-time gait video recording after training of mice from each group,and data were collected and analyzed with CatWalk XT Version 10.6. Results :Compared with sham group,in model group,the sciatic nerve function index(SFI),print length,print width,print,area,the max contact area,stand,single stance,max contact max intensity,mean intensity was significantly decreased on the PSNL left hind limb(P〈0.01);OMT treatment significantly reversed the above nine parameters on the PSNL left hind limb(P〈0.05);compared with sham group,in model group,print length,print width,print,area,the max contact area was significantly increased on the right front limb(P〈0.01);OMT treatment significantly decreased the above four indicators of the right front limb of PSNL mice(P〈0.05);Parameters related to physical coordination,such as stride length,swing,body speed and regularity index were not changed on the surgical left hind limb. Conclusion :OMT can significantly improve and adjust the gait behavior of mice with neuropathic pain.
