摘  要:目的初步探讨氧化苦参碱对神经病理性疼痛大鼠实时步态行为学改善作用。方法将50只大鼠按随机数字表方法分为假手术组、模型组、氧化苦参碱组(105 mg/kg)、硝苯地平组(8 mg/kg)和氧化苦参碱(105 mg/kg)与硝苯地平(8 mg/kg)联合给药组各10只。利用L5、L6脊神经结扎(SNL)法,建立神经病理性疼痛模型,各组大鼠经训练后分别进行实时步态行为录像,进行数据采集分析。结果身体协调性相关指标Regularity Index、Swing中,各组未见显著性差异(P>0.05);大鼠足印压力的变化中,模型组大鼠在行进过程中,结扎侧左后足(LH)足印明显变小,而结扎对侧的右前足(RF)足印变亮,给予氧化苦参碱后的SNL大鼠与模型组相比,行进时的LH足印增大,RF足印变小变暗;坐骨神经功能指数中,与假手术组比较,模型组SNL大鼠的坐骨神经功能指数显著下调(P<0.01);氧化苦参碱、硝苯地平、氧化苦参碱与硝苯地平联合给药分别使SNL大鼠的坐骨神经功能指数显著改善(P<0.01);四足足印相关参数指标中,与假手术组比较,模型组的LH足印长度、足印宽度、最大接触时间的最大强度、站立时间4项指标均显著下调,RF足的足印宽度显著上调,氧化苦参碱与硝苯地平联合给药使模型组足印长度指标在LH足显著上调,RF足显著下调;氧化苦参碱、硝苯地平、氧化苦参碱与硝苯地平联合给药使模型组LH足的足印宽度、最大接触时间的最大强度、站立时间3项指标显著上调(P<0.01)。结论氧化苦参碱能显著改善神经病理性疼痛大鼠步态行为。Objective: To primarily investigate the effect of oxymatrine on the real time gait behavior of neuro- pathic pain rats. Methods: fifty rats were randomly divided into sham-operation group ,the model group, OMT group(105 mg/kg),Nifedipine(Nif) group(8 mg/kg) and OMT(105 mg/kg)+Nif(8 mg/kg) group, 10 mice in each group. Neuropathic pain model was established by spinal nerve ligation (SNL) on the left side L5 and L6 of rats. Real-time gait video was recorded after training,and data was collected and analyzed with Catwalk XT version 10.6. Results: No significant differences of regularity index and swing were found in any groups. In the model group ,when rats walking,the foot prints were noticeably smaller on left hindpaw (LH), but were brighten up on right forepaw(RF),while OMT treatment significantly reversed this phenomenon- the foot prints were brighten up on LH and getting smaller on RF. Compared with the sham group,the sciatic nerve function index (SFI) in the model group was significantly decreased. OMT, Nif and OMT+Nif treatment significantly reversed the SFI parameter. Compared with the sham group ,print length ,print width ,max contact max intensity and stand in the model group were significantly decreased on the LH and print width was significantly increased on the RF. OMT+Nif treatment significantly increased print length on the LH and decreased it on the RF. OMT,Nif and OMT+Nif treatment significantly increased print width,max contact max intensity and Stand on LH. Conclusion: OMT can significantly improve the gait behavior of rats with neuropathic pain.
