三阳清解液乙状结肠滴注对内毒素诱导发热模型大鼠体温及血清IL-6、MIP-1含量的影响  被引量:5

摘  要:目的观察三阳清解液乙状结肠滴注对内毒素(LPS)诱导发热模型大鼠退热作用机制。方法48只大鼠随机分为6组,空白组、模型组、对乙酰氨基酚组、三阳清解液高剂量组、三阳清解液中剂量组、三阳清解液低剂量组,除空白组外,其他各组腹腔注射LPS(20μg/kg)建立大鼠发热模型,观察用药后每小时的体温变化,共6.5 h,用ELISA法测定血清中白细胞介素-6(IL-6)、巨噬细胞炎症蛋白-1(MIP-1)的含量。结果三阳清解液能显著降低LPS发热模型大鼠的体温,且量效关系较显著;三阳清解液各剂量组能显著抑制发热模型大鼠血清中IL-6、MIP-1的水平。结论三阳清解液对LPS诱导的发热大鼠模型有良好的解热作用,且有较好的剂量依赖性,其解热效果可能与其抑制发热大鼠血清中IL-6、MIP-1的含量相关。Objective: To study the autipyretic effect of sigmoid instillation of Sanyang Qingjie Decoction on rat model with endotoxin (LPS)-induced fever. Methods: 48 rats were randomly divided into six groups: the control group, the model group, acetaminophen group, high dose group of Sanyang Qingjie Decoction, middle dose group of Sanyang Qingjie Decoction,low dose group of Sanyang Qingjie Decoction. The rat model of fever was established by intraperitoneal injection of LPS (20 μg/kg), except the control group. The influence of Sanyang Qingjie Decoction on body temperature of model rats were observed every hour,lasting 6.5 hours. The levels of serum IL-6, MIP-1 in rat were detected by enzyme-linked immunosorbent assay. Results: Sanyang Qingjie Decoction could significantly reduce the body temperature of LPS-induced rats,and the dose-effect relationship was very significant;Sanyang Qingjie Decoction in each group could significantly inhibit the levels of serum IL-6 and MIP-1. Conclusion: Sanyang Qingjie Decoction has a good antipyretic effect on rat model with LPS-induced fever,and shows a better close dependence. Its mechanism may be related to inhibition of IL-6 and MIP-1 in the serum.
