摘  要:为了研究2013—2015年福建省猪繁殖与呼吸综合征病毒(PRRSV)的流行特点及变异规律,收集福建多个地区的26株PRRSV分离株,应用RT-PCR方法对其ORF3基因进行克隆及序列分析,从而了解福建地区PRRSV的遗传变异性。结果表明,26株毒株中24株为美洲型毒株(NAgenotype,type2),2株为欧洲型毒株(EUgenotype,type 1)。26株PRRSVORF3核苷酸同源性为63.1%~100%,与VR2332的同源性为64.8%~99.5%,与HP-PRRSV代表毒株JXA1、WuH1和TJ同源性为63.7%~99.8%,与NADC30同源性为66.1%~95.8%,与欧洲代表毒株LV的同源性为63.6%~90.9%。24株美洲型ORF3遗传进化树表明,福建地区PRRSV出现了2个新的亚群,3亚群(2株处于独立的分支)和4亚群(3株为类NADC30PRRSV)。欧洲型PRRSVORF3遗传进化树表明,福建地区流行的毒株为泛亚1型毒株。综上所述,福建地区PRRSV存在2种血清型毒株,美洲型流行毒株呈现遗传变异多样性。In order to research the molecular biological characteristics and variation rule of prevalent porcine reproductive and respiratory syndrome virus (PRRSV) in Fujian province during 2013-- 2015. Twenty-six PRRSV isolates from Fujian were collected, ORF3 genes were cloned and sequenced by RT-PCR in order to understand genetic variability of PRRSV in Fujian province. The result showed that, among 26 isolates 24 belonged to the North American genotype (type 2 PRRSV) ,2 isolates belonged to the European genotype (type 1 PRRSV). The nucleotide identity among twenty-six isolates ranged from 63. 1%-100%, shared 64. 8% -99. 5% homology with VR2332;63. 7%-99. 8% with JXAI,WUH1 and TJ;66. 1%-95. 8%. with NADC30 and 63. 6%- 90.9% with LV. The phylogenetic tree analysis showed that there were two new suhgenotypes in these Fujian PRRSV strains,subgroup Ⅲ (two isolates formed a new subgenotype ) and subgroup Ⅳ (three isolates formed NADC30 PRRSV). The genetic evolution tree of European type PRRSV ORF3 showed that the prevalent strain in Fujian was Fan Asian type 1 strain. These results showed that type land type 2 coexisted in the Fujian province and type 2 PRRSV have been shown a remarkable genetic diversity.
