猪圆环病毒2型衣壳蛋白Loops结构及病毒样颗粒制备的研究进展

摘　　要：病毒样颗粒（virus—like particles，VLPs）是某种病毒的1种或几种结构蛋白在体外组装而成，由于VLPs不包含病毒基因组，在宿主细胞q-不能自我复制，因此VLPs在亚单位疫苗应用中具有较高的生物安全性。自猪圆环病毒2型（poreine circovirus type2，PCV2）在世界范围内流行以来，已经给全球养猪业带来巨大经济损失，因此PCV2的致病机理和疫苗研究具有重要现实意义。PCV2Cap蛋白（capsid protein）是病毒唯一的衣壳蛋白，也是主要的免疫原性蛋白，60个Cap蛋白亚基在体外能自行组装成PCV2VLPs，该VLPs因与天然病毒粒子形态相似并保留了病毒完整的构象表位，已经在PCV2组装和侵染机理研究、亚单位疫苗的研发及其相关性系统疾病（PCV2-systemic disease，PCV2-SD）的预防研究中得到广泛应用。本文主要对PCV2Cap蛋白的Loops结构及其VLPs的制备研究进展进行阐述，为PCV2侵染机理与亚单位疫苗的研发等提供参考。Virus-like particles （VLPs） are spontaneously assembled by one or several viral structure proteins in vitro. VLPs have no characteristics of self-replication in host cells due to absence of viral genomes,and have a stronger safety profile in subunit vaccine application. Porcine circovirus type 2（PCV2） is a common virus of pigs found throughout the world,causing huge economic loss for the worldwide swine industry, the pathogenic mechanism and effective vaccines against PCV2 have important significance. As the only structural protein of viral capsid, PCV2 capsid protein （Cap） is major immunogenic protein,and has become the major antigen for development of vaccines. The 60 expressed Cap protein can self-assemble into VLPs, mimicking the live virus with the complete comformational epitope on the surface,it has been widely used for the development of PCV2 subunit vaccines and the prevention of PCV2-systemic disease （PCV2-SD） and the PCV2 infection mechanism. Here we review the structure of PCV2 Cap Loops and the PCV2 VLPs produced in different systems, which provide references for the study of PCV2 infection mechanism and further development in PCV2 subunit vaccines.
