摘  要:目的探讨菟丝子黄酮基于Wnt/β-catenin信号通路干预去卵巢大鼠骨代谢的作用机制。方法将100只健康大鼠随机分为假手术组、模型组、实验组(菟丝子黄酮高剂量组、中剂量组及低剂量组),每组20只。模型组和实验组在输卵管壶腹处进行结扎,然后切除两侧卵巢;假手术组仅行开腹手术。实验组给予相应剂量的菟丝子黄酮混悬液灌胃,假手术组和模型组给予等量的生理盐水灌胃。12周造模结束后,处死大鼠并取材。测定五组大鼠全身、右侧股骨近端2 cm和4~6腰椎骨的骨密度及血清β-链蛋白(β-catenin)、骨保护素(OPG)、细胞因子Kb受体活化因子配体(RANKL)、DKK1水平。分析血清β-catenin水平与OPG、RANKL的相关性。结果假手术组,高、中、低菟丝子黄酮剂量实验组在全身、股骨近端2 cm及4~6腰椎骨密度上均高于模型组(P<0.05);假手术组、高中低菟丝子黄酮剂量实验组的血清β-catenin、OPG水平高于模型组(P<0.05),假手术组、高中低菟丝子黄酮剂量实验组的血清RANKL、DKK1水平低于模型组(P<0.05);高、中、低菟丝子黄酮剂量实验组的血清β-catenin水平均与OPG水平呈正相关(P<0.05),与RANKL、DKK1水平呈负相关(P<0.05)。结论菟丝子黄酮能够调节去卵巢大鼠Wnt/β-catenin信号通路相关因子β-catenin、OPG、RANKL、DKK1的水平,从而增加骨密度,改善骨质疏松症状。Objective To explore the dodder flavonoids based on Wnt/ beta catenin signaling pathway to ovarian mechanism of action of bone metabolism in rats. Methods 100 healthy rats were randomly divided into control group,model group,the experimental group (dodder fla-vonoids high dose group,middle dose group and low dose group),20 in each group. Model group and experimental group in the fallopian tube am-pulla ligation,and then cut off both ovaries;sham operation group only laparotomy. Experimental group were given corresponding dose of dodder flavonoids suspension liquid lavage,control group and model group was given the same amount of saline lavage. After 12 weeks the mold was made,and the rats were put to death for materials. 2 cm on the right side of proximal femur,and 4 ~ 6 cm bone mineral density and serum beta- catenin,OPG and RANKL,DKK1 level of all five groups were detected. Results Whole body,2 cm proximal femur,and 4 ~ 6 cm bone min-eral density were higher than those of the model group (P 〈 0. 05). The beta - catenin,serum OPG levels were higher than those of the model group and control group (P 〈 0. 05),serum RANKL,DKK1 level were lower than those of the model group (P 〈 0. 05). The serum beta cate-nin and OPG levels the in experimental group were positively correlated (P 〈 0. 05),and negatively correlated with RANKL,DKK1 level (P 〈0. 05). Conclusion Dodder flavonoids can adjust the ovaries rats Wnt/ beta - catenin signaling pathway related factor beta catenin,OPG and RANKL,DKK1 levels,thus increase bone mineral density,improve the symptoms of osteoporosis.
