摘 要:目的观察和比较比索洛尔联用中药大黄治疗前后大鼠血压、血中一氧化氮(NO)、一氧化氮合酶(eNOS)和内皮素(ET)的时间生物学特征的变化。方法健康雄性SD大鼠120只,体重250克左右,随机分为3个大组,随机分为对照组(对照组给予安慰剂)(组Ⅰ)、灌服比索洛尔组(3 mg·kg^(-1)·d^(-1))(组Ⅱ)及灌服比索洛尔(3mg·kg^(-1)·d^(-1))和大黄组(300 mg·kg^(-1)·d^(-1))(组Ⅲ)。每大组又随机分为4小组。服药前1 d,分别于同日内不同时间点(02∶00,08∶00,14∶00,20∶00)用鼠尾血压仪测3大组中各1小组大鼠收缩压。给予上述喂养4周后,于4周末最后1 d内不同时间点,02∶00,08∶00,14∶00,20∶00,用鼠尾血压仪测3大组中各1小组大鼠收缩压并处死,分别留取血浆、血清。分别采用放免法、比色法和硝酸还原法检测血中ET-1、eNOS、NO的水平。结果与对照组相比,治疗组大鼠的收缩压、ET-1水平均下降,血中NO、eNOS水平上升,均以组Ⅲ较明显,差异均有统计学意义(P<0.05);治疗组大鼠的血压及内皮系统的昼夜节律特征均有明显改变,也以组Ⅲ表现较明显,差异均有统计学意义(P<0.05)。结论比索洛尔联合大黄具有协同作用,能更明显降低血压,同时对于维持血压昼夜节律、改善内皮系统功能有明显影响,提示中西医结合治疗高血压的方式可能更有效的控制血压。Objective To investigate the effects of bisoprolol combined with Dahuang on the changes of circadian rhythm of blood pressure( BP),the levels of nitric oxide( NO),nitric-oxide synthase( eNOS) and endothelin( ET) in experimental rats. Methods A total of 120 healthy male SD rats weighted about 250 grams were randomly divided into three groups,control group( groupⅠ),group Ⅱ and Dahuang group( group Ⅲ),moreover,each group was randomly redivided into 4 subgroups. The rats in groupⅠwere given placebo by gavage,and the rats in group Ⅱ were given bisoprolol 3 mg·kg^(-1)·d^(-1) by gavage,however,the rats in group Ⅲ,on the basis of group Ⅱ,were given Dahuang 300 mg·kg^(-1)·d^(-1). On 1 day before medication,the levels of systolic pressure in different time pionts( 02 ∶ 00,08 ∶ 00,14 ∶ 00,20 ∶ 00) were detected by rat-tail manometry,respectively in one subgroup of every group. After 4-week feeding,the levels of systolic pressure were detected by rat-tail manometry,respectively in one subgroup of every group,then all the rats were sacrificed,and the serum levels of ET-1,eNOS,NO were detected by radioimmunity method,colorimetric method and nitric acid reduction method,respectively.Results As compared with those in groupⅠ,the levels of systolic pressure and ET-1 were decreased in group Ⅱ and groupⅢ,however,the serum levels of NO and eNOS were increased in the two groups,moreover,the degree of increase or decrease in group Ⅲ was more obvious than that in group Ⅱ( P 〈0. 05). In addition the circadian rhythm characteristics of blood pressure and endothelial system were significantly changed in group Ⅱ and group Ⅲ,moreover,the changes in group Ⅲ were more obvious than those in group Ⅱ( P 〈0. 05). Conclusion The bisoprolol combined with Dahuang has synergistic effects on the serum levels of NO,eNOS and ET,which can decrease obviously blood pressure,keep circadian rhythm of blood pressure and improve endothelial system function,which
