摘　　要：[目的]研究乌鳢的肌肉、肝脏、肾脏、尾鳍、鳞片、肠、鳃盖、鳃等组织的DNA提取。[方法]利用酚-氯仿法提取乌鳢的肌肉、肝脏、肾脏、尾鳍、鳞片、肠、鳃盖、鳃等组织的DNA,采用紫外分光光度计法、琼脂糖凝胶电泳法和PCR扩增法检测提取的DNA质量。[结果]结果表明,不同组织的DNA含量不同(0.370~0.975μg/μL),其中肌肉、肝脏、肾脏和肠等组织DNA质量较高,而尾鳍、鳞片、鳃和鳃盖等组织DNA质量较差。[结论]乌鳢的肌肉、肝脏、肾脏和肠等组织提取的DNA可用于后续的分子生物学研究。[Objective] To research DNA extraction from the muscles,liver, kidney, caudal fins, scales, intestines, operculum and gills of Ophiocephalus argus Cantor. [Methods]Genomic DNA from the muscles,liver,kidney,caudal fins,scales,intestines,operculum and gills of Ophiocephahts argus Cantor was extracted by phenol-chloroform.DNA concentration of various tissues of Ophiocephalus argus Cantor calculated by ultraviolet spectrophotometer tests and agarose gel electrophoresis.Mitochondrial DNA control region sequence of Ophiocephalus argus Cantor was amplified by PCR.[Results] The DNA concentration of different tissues of Ophiocephdus argus Cantor ranged from 0.370 to 0.975 μg/μL.The quality of DNA in muscle ,liver, kidney and intestine was higher than that in caudal fin, scales, gills and gill cover. [Conclusion]DNA extracted from the muscle ,liver,kidney and intestine of Ophicephalus argus Cantor can be used in subsequent molecular biology research.
