摘 要:目的:观察硝菔通结方对功能性便秘大鼠肾脏以及结肠组织水通道蛋白的影响,从现代医学肾脏调节水液代谢的角度探讨中医"肾主水司二便"的科学内涵,揭示硝菔通结方从肾论治便秘的作用机制。方法:48只成年雄性SD大鼠,采用随机法分为空白组(8只)和造模组(40只),造模组灌胃复方地芬诺酯混悬液6周复制便秘模型后随机分为模型组(FC组)、硝菔通结方高、中、低剂量组、阳性药物对照组(苁蓉通便口服液组,简称口服液组)。各治疗组给予不同剂量的硝菔通结方、阳性药物对照组给予苁蓉通便口服液(后简称口服液组)灌胃1个月。观察各组大鼠的一般情况,采用ELISA法检测各组大鼠血清中精氨酸加压素(AVP)、环磷酸腺苷(c AMP)的含量,Western-Blot法检测肾脏组织中AQP2以及AQP3的表达,结肠组织中AQP3的表达。结果:与空白组比较,模型组大鼠粪便量少、质地干硬,血清中AVP、c AMP的含量升高(P〈0.05),肾脏组织中AQP2以及AQP3的表达明显升高(P〈0.01),结肠组织中AQP3的表达明显升高(P〈0.01)。与模型组相比,各剂量硝菔通结方组以及口服液组大鼠粪便量增加且质地稀软,血清中AVP、c AMP的含量降低(P〈0.05),肾脏组织中AQP2以及AQP3的表达明显降低(P〈0.01),结肠组织中AQP3的表达明显降低(P〈0.01)。结论:肾脏能通过调节水通道蛋白而影响肠道的水液代谢,这可能是中医"肾主水司二便"的科学内涵之一;硝菔通结方可通过影响肾脏组织的水通道蛋白而纠正肠道水液代谢紊乱,从而对功能性便秘发挥治疗作用。Objective:To study the aquaporin water channels in the kindey and the colon of the treatment of functional constipation in Xiaofu Tongjie Formula. From the view of the kindney adjusting water metabolism,we discussed the scientific connotation of kindney controlling water metabolism and governing defecation to reveal the mechanism of treating functional constipation from the kindney. Methods:Forty-eight SD rats were randomly divided into 6 groups: blank control group,functional constipation model group,Xiaofu Tongjie Formula high,medium and low dose groups,positive drug group,8 rats in each group. Model group and other treatment groups were given compound diphenoxylate 15 mg/( kg·d)by gavage for 6 weeks,and then treated with Xiaofu Tongjie Formula. Positive drug group was treated with Congrong Tongbian oral liquid. All for more than one months. We observed the general condition of each group. The protein expression of the arginine vasopressin( AVP) and the cyclic adenosine monophosphate( c AMP) were detected by enzyme linked immunosorbent assay( ELISA). The protein expressions of AQP2 and AQP3 in kidney and AQP3 in colon were detected by Western bolt. Result:Compared with the blank control group,the rats of model group had less feces and texture was hard. AVP and c AMP contents were increased( P 〈 0. 05). Levels of AQP3 relative expression levels were significantly increased( P 〈 0. 01). After giving Xiaofu Tongjie Formula,the rats of treatment groups had more feces and texture was loose. AVP and c AMP contents were decreased( P 〈 0. 05). AQP3 expression levels reduced compared with that of the model group( P 〈 0. 01). Conclusions:The kindey can affect water metabolism disorder in colon by adjusting aquaporins. This may be one of the scientific connotation of nursing functional constipation based on the kindney controlling water metabolism and governing. Xiaofu Tongjie Formula may affect the aquaporin water channels in the kidney to regulatingbody’s water metabo
