摘 要:目的:研究西黄丸对结肠癌耐药细胞株HCT-116/L及HCT-8/V的体外逆转作用。方法:血清药理方法制备西黄丸含药血清。用细胞毒性试验CCK8法、Western Blotting,RT-PCR方法研究西黄丸含药血清对大肠癌耐药细胞株的逆转作用及可能的分子机制。结果:西黄丸含药血清联合不同浓度长春新碱(VCR)与奥沙利铂(L-OHP)处理48 h后,对HCT-8/V细胞和HCT-116/L细胞的逆转倍数分别为2.58和2.36;西黄丸含药血清处理HCT-8/V细胞和HCT-116/L细胞48 h后,PKCα、Nrf2、MRP1、MRP2、GSTp1基因及蛋白,除HCT8/V细胞株中MRP2基因(6倍7 d组)上调外,表达均下调,差异有统计学意义(P〈0.05)。结论:西黄丸含药血清可体外逆转结肠癌耐药细胞株HCT-8/V和HCT-116/L的耐药性,可能与下调耐药基因mRNA和蛋白的表达水平有关。Objective: To investigate the reversal effect of Xihuang Pill on human colon cancer cell lines HCT-116/L and HCT-8/V in vitro. Method: Serum pharmacological method was used to prepare Xihuang Pill-containing serum.The reversal effect and possible molecular mechanism of drug-containing serum on colorectal cancer cell line were tested by cytotoxicity test CCK8,Western Blotting and RT-PCR. Result: The reversal multiples of HCT-8/V cells and HCT-116/L cells were 2. 58 and 2. 36. PKC,Nrf2,MRP1,MRP2 and GSTp1 gene and protein expressions were decreased after HCT-8/V cells and HCT-116/L cells were treated with Xihuang Pill-containing serum for 48 h( P 〈 0. 05). In addition,the expression of MRP2 gene was up-regulated in HCT-8/V cells( six times and seven days). Conclusion:The drug-containing serum of Xihuang Pill could reverse the drug resistance of HCT-116/L and HCT-8/V in vitro,which may be related to the down-regulation of mRNA and protein expression which is related to multidrug resistance.
