摘 要:目的研究洋金花对单胺氧化酶B(MAO-B)的抑制效果及抑制类型。方法采用醇提水沉法对洋金花不同部位进行提取,获得提取物,并计算得率。利用分光光度法研究了洋金花不同部位提取物对MAO-B的抑制作用。对花提取物抑制数据进行分析,求得花提取物对MAO-B的半数抑制浓度IC_(50)值。进一步对花提取物抑制MAO-B的动力学进行考察,根据双倒数曲线结果判定抑制机制。结果洋金花提取物对MAO-B有抑制作用,花部位提取物抑制效果最强,浓度为1 mg/ml时,对MAO-B的抑制率为(55.74±1.5)%,半数抑制浓度IC_(50)为0.75 mg/ml。抑制动力学研究结果表明,洋金花提取物对MAO-B的抑制作用属于混合型抑制。结论洋金花提取物对MAO-B具有明显抑制作用,属于混合性抑制,这可能是洋金花对PD具治疗作用的机制之一。Objective To study the inhibitory effect and types of Datura metel L. extract on monoamine oxidase B(MAO)-B. Methods Alcohol extraction water sedimentation method was used to obtain fractions from different parts of Datura metel L., and the yields of the extracts were calculated. Spectrophotometry was used to study the inhibitory effect of different parts of Datura metel L. on MAO-B. The inhibition data of flower extract were analyzed, and the IC50 value of half inhibitory concentration of flower extract to MAO-B was obtained. The kinetics of MAO-B inhibition was further investigated, and the inhibition mechanism was determined according to the results of the double reciprocal curve. Results The extracts of Datura metel L. had the inhibitory effect on MAO-B, and the extracts of flower parts had the strongest inhibitory effect. The inhibitory rate of MAO-B was(55.74±1.5)% at the concentration of 1 mg/ml and the IC50 was 0.75 mg/ml. Inhibition kinetics showed that the inhibitory effect of Datura metel L. on MAO-B was mixed type inhibition. Conclusion Datura metel L. has obvious inhibitory effect on MAO-B, which belongs to the mixed inhibition. It may be one of the mechanisms of Datura metel L. in healing PD.
