摘  要:目前马骨关节炎(OA)的临床诊断尚缺少有效的生物标志物(BM)。为探讨马关节液Ⅱ型胶原羧基端端肽(CTX-Ⅱ)的浓度与OA发病程度的关系。试验选用13匹蒙古马,随机分为造模组(n=8)和对照组(n=5),在马左侧腕关节内注入2 mL两性霉素B(25 mg/mL)诱导OA,对照组马匹注入等量PBS作为对照,于造模后每周采集一次关节液,直至第9周。ELISA检测马关节液中的CTX-Ⅱ浓度。结果显示,与对照组相比,造模组从第1周开始,关节液中CTX-Ⅱ的浓度增加,从第2周开始显著增加(P<0.05),第4周开始极显著增加(P<0.01)并且保持增加的趋势到第9周。造模组马匹第3周开始出现不同程度跛行。试验结果表明,CTX-Ⅱ浓度的升高在跛行前就已经显著升高,推测CTX-Ⅱ可以作为马关节炎BM,为以后建立马OA的快速临床诊断技术提供理论依据。There are no effective biomarkers (BM) for the diagnosis of equine osteoarthritis currently. To investigate the rela- tionships between the concentration of C-terminal telopeptide of type Ⅱ collagen ( CTX -Ⅱ ) and the degree of OA in synovial fluid, a total of 13 Mongolia horses were randomly divided into model group (n = 8 ) which was injected with 2ml amphotericin B (25 mg/ ml) into the left wrist to induce osteoarthritis and control group ( n = 5 ) which was administrated with the same dose of PBS. Then, the synovial fluid was collected once a week after modeling until the 9th week to detect the concentration of CTX - Ⅱ by ELISA. The results indicated that the concentration of CTX - Ⅱ in the model group changed in a growing trend within 9 weeks compared to the control group, which rose sharply from the second week ( P 〈0. 05) and increased remarkably at the 4th week ( P 〈0. 01 ). The horses in the model group appeared limping symptom in vary degres in the third week. The conclusion is that the growing concentration of CTX - Ⅱ has been significantly increased before lameness so that CTX - Ⅱ could be used as a biomarker of equine OA for the establishment of rapid clinical diagnosis of OA.
