摘 要:目的确定柴芍安神解郁颗粒水提液的纯化工艺。方法以芍药苷和斯皮诺素的相对保留率及除杂率的综合评分为评价指标,采用单因素试验比较离心沉淀法、乙醇沉淀法、澄清剂法(壳聚糖澄清剂、果汁101澄清剂、ZTC1+1澄清剂)等不同纯化方法对提取液纯化效果的影响;采用三因素四水平正交试验设计对乙醇沉淀法进行纯化工艺参数优选。结果乙醇沉淀法的纯化效果优于离心法和澄清剂法,最佳纯化工艺为:药液浓度为0.5 g·mL^(-1),加乙醇至药液含醇量为70%,静置24 h。结论优选的乙醇沉淀工艺可用于柴芍安神解郁颗粒水提液的纯化,该方法合理、稳定可行。OBJECTIVE To optimize the purification technology of the water extract of Chaishao Anshen Jieyu Granules (CAJG). METHODS The single factor tests were adopted to eompre the purification effects of ethanol precipitation method, centrifugation method and clarifier method( including chitosan clarifier,juice 101 clarifier, ZTC1 + 1 clarifier) on the water extract of CAJG by using the comprehensive scores of the relative retention rate of contents of peoniflorin, spinosin and the yield of dry extract as the evaluation index. Three factors and four levels orthogonal design was used to optimize the process conditions of ethanol precipitation method. RESULTS The purification effect of ethanol precipitation method was better than the method of centrifugation and clarifier, the optimal purification process was as following:the concentration of the water extract was 0.5 g. mL-l , the amount of alcohol was 70%, and keep static 24 h. CONCLUSION The ethanol precipitation method can be used for purifying the water extract of the CAJG, and the process is reasonable, stable and feasible.
