摘 要:目的研究白桦脂醇对阿霉素(ADMiamycin,ADM)所致大鼠心脏损害的保护作用。方法将60只大鼠随机分成正常对照组、ADM模型组和白桦脂醇治疗组。苏木素-伊红(HE)染色检测心肌组织病理,测定比较各组大鼠心率(heart rate,HR)、左室收缩压(left ventricular systolic pressure,LVSP)、左室舒张末压(left ventricular end diastolic pressure,LVEDP)、左室内压最大上升速率(+dp/d tmax)、左室内压最大下降速率(-dp/d tmin)、血清肌钙蛋白I(cardiac troponin I,c Tn I)、肌酸激酶(creatine kinase,CK)、超敏C-反应蛋白(high-sensitivity C-reactive protein,Hs-CRP)、B型利钠肽(B-type natriuretic peptide,BNP)、乳酸脱氢酶(lactate dehydrogenase,LDH)、血浆超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)水平。免疫染色组化法测定并比较各组大鼠心肌细胞凋亡率、B细胞淋巴瘤/白血病基因-2(B-cell lymphonma/leukemia gene 2,Bcl-2)和Bcl-2相关基因X(Bcl-2 Associated X protein,Bax)表达。采用传统阳性染色细胞计数的方法对各组大鼠染色阳性细胞进行计数评分。结果与对照组和白桦脂醇组相比,模型组大鼠心功能指标降低,血清c Tn I、CK、Hs-CRP、BNP、LDH和MDA水平升高,SOD活性降低,差异有统计学意义(P<0.05)。对照组和白桦脂醇组大鼠心肌细胞凋亡率与细胞Bcl-2表达均低于模型组,差异有统计学意义(P<0.05),心肌细胞Bax表达高于模型组(P<0.05)。模型组阳性细胞染色评分均高于对照组和白桦脂醇组。结论白桦脂醇对阿霉素诱导的大鼠心肌损伤具有保护作用,其机制可能白桦脂醇抑制氧化应激相关。Objective Objective To explore the protective effects of botulin on the cardiac injury induced by ADM in rats. Methods Total 60 rats were randomly divided into normal control group, ADM model group and botulin treatment gourp. Myocardial pathology were examined by staining of HE. The level of cTnI, CK, Hs-CRP, BNP, LDH, SOD, MDA and the impact of cardiac function including HR, LVSP, LVEDP, + dp/d tmax and - de/d train were examined. The immunohistoehemieal method was used to examine the cardiae myocyte apoptosis rate and the expression of bcl-2 and bax. Traditional positive staining cells counting method was used to calculate the positive staining cells between different rat groups. Results Compared with normal control group and botulin group, the index of cardiac function in ADM model group was decreased, the level of cTnI, CK, Hs-CRP, BNP, LDH, MDA were significantly increased and the level of SOD was decreased (P 〈 0. 05 ). The myocyte apoptosis rate and bc1-2 expression were significantly lower than those in model group ( P 〈 0. 05 ) , while the bax expression was significantly higher (P 〈 0. 05 ). The positive staining cells counting in model group was significantly higher when compared with normal control group and botulin group (P 〈 0. 05). Conclusion The botulin has the protective effects on the cardiac injury induced by ADM the mechanism may be related with the inhibition of oxidative stress induced by botulin.
