摘  要:探究环境因子对土壤微生物反硝化气体产生的影响,选取培养温度、氧气含量、亚硝酸盐含量、葡萄糖含量、p H值、Cu^(2+)含量6个因素,分别设计3个水平,设计7因素3水平一共18组的正交实验,对土壤有机物含量和培养条件进行调控,在密闭环境下进行培养,同时监测顶空气体中反硝化气体含量,连续监测10 d,随后对土壤中氧化亚氮还原酶基因nos Z进行定量PCR检测。培养实验显示在18组条件中一共出现了3种的反硝化气体产生类型,分别为快速产N_2类型、N_2O累积类型和无气体产生类型,表明环境条件的差异导致了土壤反硝化微生物不同的产气过程。对定量PCR的方差分析显示单个环境因素并没有对nos Z基因拷贝数产生显著影响,表明反硝化气体产生的差异并不是通过影响nos Z基因拷贝数而产生的。This study aimed to explore the effects of environmental factors on soil microbial denitrification and gas production.Six factors,including culture temperature,oxygen content,nitrite content,glucose content,p H value,and Cu2+content were used for designing total 18 or thogonal experiments for seven factors at three levels.Those experiments were performed within closed environment to regulate and control soil organic contents and culture conditions.The content of denitrifying gas in the headspace was monitored once a day for 10 days,as well as nitrous oxide reductase genes(nos Z)copies number was quantified by PCR after the anaerobic incubation.The results showed that there were three gas production patterns in 18 experiment groups.These patterns,including rapid completion of denitrification,accumulation of N2O and no denitrification,showed different environmental conditions could cause different gas production process.The q PCR results showed that each single environmental factor didn’t significantly impact nos Z gene copy numbers,which meant the differences in denitrifying gases production were not caused by the influence in nos Z gene copy numbers
