摘 要:目的探讨孕期砷暴露对子代小鼠成年后体重、各主要器官重量和脏器系数等指标的跨代遗传效应。方法 SPF级CD1妊娠小鼠按体重随机分为对照组和亚砷酸钠染毒组(85 mg/L),每组6只,于孕期第8至18天进行自由饮水方式染毒。F1~F3代仔鼠出生后正常饲养至13周,称重,计算肾脏、脾脏、胰腺、肝脏及肺脏脏器系数。结果与对照组相比,染毒组F1代小鼠体重、肾脏、胰腺和肝脏重量及其脏器系数均明显升高(P<0.05),脾脏重量及其脏器系数明显降低(P<0.05)。按性别分组后发现,染毒组F1代雄性仔鼠体重、肾脏、胰腺和肝脏重量及其脏器系数均高于对照组(P<0.05),脾脏和肺脏脏器系数均低于对照组(P<0.05);染毒组F1代雌性仔鼠肝脏重量及脏器系数高于对照组(P<0.05),脾脏系数低于对照组(P<0.05)。染毒组F3代雄鼠肾脏重量及脏器系数明显小于对照组(P<0.05),染毒组F3代雌鼠肝脏重量及脏器系数明显大于对照组(P<0.05)。结论宫内砷暴露可以引起F1代小鼠体重及不同脏器重量和脏器系数发生改变,且部分效应在F3代也得到了继承,表现出一定的跨代遗传效应。Objective To investigate the transgenerational genetic effects of in utero arsenic exposure on body weight, organ weight and organ coefficient of mice offspring. Methods SPF-class pregnant CD1 mice were randomly divided into control group and arsenic-exposed group. Six mice were assigned in each group.During the gestation day 8-18, mice in exposed group were administrated with drinking water containing sodium arsenite(85 mg/L). Thirteen weeks after birth, F1-F3 mice offspring were weighed. Organs of each mouse, including kidney, spleen, pancreas, liver and lung,were harvest and weighed and organ coefficients were calculated. Results Compared with the control group, the body weight, kidney, pancreas and liver weight and corresponding organ coefficients of F1 mice offspring in arsenic-exposed group were significantly higher( P 0.05). However,spleen weight and its organ coefficient were significantly lower(P 0.05). For F1 male offspring, the body weight, kidney,pancreas and liver weight and corresponding organ coefficients in arsenic-exposed group were higher than these in control group(P〈0.05); Lung and spleen organ coefficients were lower than these in control group(P〈0.05). For F1 female offspring,higher liver weight and organ coefficient and lower spleen coefficient were observed in arsenic-exposed group( P 0.05).Compared with control group, there were lower kidney weight and organ coefficient of F3 male offspring in arsenic-exposed group(P 0.05), and higher liver weight and organ coefficient of F3 female offspring in arsenic-exposed group(P 0.05).Conclusion The body weight and different organs weight and organ coefficients of F1 offspring were changed due to in utero arsenic exposure. Some of the changes can be observed in F3 offspring, which indicates a transgenerational genetic effect of in utero arsenic exposure.
