摘 要:目的观察脐带间充质干细胞(UCMSC)治疗对放射伤树鼩外周血和脾淋巴细胞表达Th1/Th2类细胞因子的影响。方法成年雄性树鼩随机分成模型组、治疗组,每组15只,两组均接受全身一次性4.5 Gy电离辐照,然后治疗组经股静脉输注同种异体UCMSC(3×10~7个/kg),而模型组则给予同体积生理盐水,均1次/w,连续治疗4次。末次MSC治疗后1 w,统计两组的存活率,实时荧光定量PCR检测两组外周血和脾淋巴细胞IL-2、IL-4、IL-6、TNF-α的m RNA表达水平。结果在末次治疗后1 w,治疗组存活率为86.677%,模型组存活率为53.33%,两组比较有显著差异(P﹤0.05);治疗组外周血和脾淋巴细胞TNF-α、IL-2表达量均低于模型组(P﹤0.05),而IL-4、IL-6表达量均高于模型组(P﹤0.05)。结论 UCMSC治疗可提高全身放射损伤树鼩的生存率,诱导淋巴细胞Th1/Th2类细胞因子表达向Th2优势化。Objective To observe the impact of umbilical cord-derived mesenchymal stem cells (UCMSCs) treatment on the expression of Th1/Th2 cytokines in peripheral and splenic lymphocytes of tree shews with radiation injury. Methods Adult male tree shrews were randomly divided into a model group and a treatment group (n=15, respectively); both groups received one-time ionizing radiation of 4.5 Gy to the whole body; and then, the treatment group received an infusion of allogeneic UCMSCs (3×10^7 cells/kg) via the femoral vein and the model group was treated with normal saline of the same volume, and both groups were treated for four consecutive times, once per week. One week after the last MSC treatment, the statistical treatment was done on the survival rate of both groups, and then the real-time quantitative polymerase chain reaction (PCR) technique was performed on the peripheral and splenic lymphocytes in both groups to obtain the mRNA expression levels of IL-2, IL-4, IL-6 and TNF-α. Results One week after the last treatment, the survival rate of the treatment group was 86.677% and that of the model group was 53.33%, presenting a significant difference (P 〈 0.05); the expression levels of TNF-α and IL-2 in the peripheral and splenic cells of the treatment group were lower than those in the model group (P 〈 0.05), while the expression levels of IL-4 and IL-6 were higher than those in the model group (P 〈 0.05). Conclusioa The UCMSCs treatment can improve the survival rate of tree shrews with radiation injury to the whole body and induce a switch of the Th1/Th2 cytokine expression in lymphocytes to the dominant Th2 cytokine expression.
