摘  要:目的观察人脐带间充质干细胞(UCMSC)对体外CD4^+和CD25^+Treg淋巴细胞的影响。方法分离培养人UCMSC和淋巴细胞,将UCMSC和淋巴细胞按不同比例分别以接触和非接触方式共培养72 h,同时设淋巴细胞单独培养为对照组。收集各组淋巴细胞后,采用流式细胞术检测CD4^+和CD25^+Treg细胞的百分比,并取各组上清液,采用ELISA法检测IL-10含量。结果当淋巴细胞与UCMSC以10∶1接触和非接触共培养时,Treg细胞比例最高,各共培养组CD4^+和CD25^+Treg细胞和IL-10含量显著高于对照组(P<0.05)。结论 UCMSC可以诱导淋巴细胞向Treg细胞的分化,其作用机制可能与UCMSC分泌的细胞因子有关。Objective To observe the effect of umbilical cord-derived mesenchymal stem cells (UCMSCs) on CD4+ CD25+ Treg cells in vitro. Methyls Human UCMSCs and lymphocytes were isolated and co-cultured in different proportions in contact and non- contact modes for 72 h; additionally, other lymphocytes were isolated and cultivated to be a control group. Lymphocytes of all groups were collected and treated with the flow cytometry to measure the percentage of CD4+ CD25+Treg cells; and then, the supernate of all groups was obtained and treated with the enzyme-linked immunosorbent assay (ELISA) method to measure the level of IL-10. Results When lymphocytes were co-cultured in contact and non-contact modes with UCMSCs in the proportion of 10:1, Treg cells accounted for the highest percentage among all ceils; the levels of CD4+ CD25+Treg ceils and IL-10 in all co-culture groups were significantly higher than those in the control group (P 〈 0.05). Conclusion UCMSCs could induce lymphocytes to differentiate into Treg cells; the mechanism of such differentiation may be related to cytokines released by UCMSCs.
