摘  要:目的探讨水母雪莲黄酮对中波紫外线(UVB)辐射人永生化角质细胞(HaCaT)损伤的影响。方法将体外培养的HaCaT细胞株随机分为两组,给予25 m J/cm^2和50 m J/cm^2剂量的UVB进行照射,然后将细胞进一步分为四组:对照组、UVB模型组、水母雪莲黄酮2 g/L组、水母雪莲黄酮8 g/L组。MTT法检测细胞增殖,酶标法检测超氧化物歧化酶(SOD)活性、谷胱甘肽(GSH)含量、细胞丙二醛(MDA)含量及过氧化氢酶(CAT)活性。结果在不同剂量紫外线辐射组中,与对照组比较,UVB模型组细胞吸光度降低,SOD活性、CAT活性及GSH含量明显下降,MDA含量明显上升,差异均有统计学意义(P<0.05)。与UVB模型组比较,水母雪莲黄酮组细胞吸光度增加,SOD活性、CAT活性及GSH含量明显上升,MDA含量明显下降,差异均有统计学意义(P<0.05)。结论水母雪莲黄酮对UVB辐射后HaCaT细胞的氧化损伤有一定的保护作用。Objective To investigate the effects of flavones derived from Saussurea medusa Maxim. for the HaCaT cells injury caused by UVB radiation. Methods The HaCaT cells line cultured in vitro was randomly divided into two groups, which were respectively given UVB irradiation at doses of 25 mJ/cm^2 and 50 mJ/cm^2. Then, the cells were further subdivided into four groups, including control group, UVB model group, 2 g/L of flavones derived from Saussurea medusa Maxim. group, and 8 g/L of flavones derived from Saussurea medusa Maxim. group. The cell proliferation was determined by MTT assay. The activities of superoxide dismutase (SOD), contents of glutathione (GSH), malondialdehyde (MDA) and activities of catalase (CAT) were detected by enzyme linked immunosorbent assay. Results In different doses of UVB ra- diation groups, compared with control group, the absorbance of UVB model group was decreased, the activities of SOD, CAT and the contents of GSH were reduced, while the contents of MDA were increased, the differences were all statistically significant (P 〈 0.05). Compared with UVB model group, the absorbance of flavones derived from Saussurea medusa Maxim. group was increased, the activities of SOD, CAT and the contents of GSH were enhanced, while the contents of MDA were decreased, the differences were all statistically significant (P 〈 0.05). Conclusion The flavones derived from Saussurea medusa Maxim. have certain protective effects for the oxidative damage of HaCaT cells after UVB radiation
