信阳土鸡禽白血病的分子生物学与形态学诊断

摘　　要：为全面了解信阳地区土鸡种群中禽白血病的分子流行病学特点,笔者对信阳地区部分规模化鸡场的土鸡（肉蛋兼用型）进行A、B亚群和J亚群禽白血病病毒（ALV）抗体检测和p27抗原检测。并用外源性ALV-A、B、J亚群的特异性序列作为引物,对部分p27阳性病例肝样品进行了PCR检测,同时进行病理组织学分析。结果显示,在调查的92份样品中,ALV-A/B抗体阳性率为5%;ALV-J抗体阳性率为36%;禽白血病p27抗原阳性率为74%。提取10份p27抗原阳性病鸡肝的总RN A,并进行PCR检测,发现ALV-A、B亚群阳性各有1例、J亚群有2例;病理组织学检查发现在肝、肾等部位可见淋巴细胞样肿瘤病灶和髓细胞瘤样病灶。本研究结果提示信阳地区土鸡群中存在着不同亚群禽白血病病毒的感染病例。In order to investigete comprehensively the characteristics of molecular epidemiology of avian leukemia in native chicken population in Xinyang,many chickens raised for meat and eggs pro- duction in some large-scale chicken farms were surveyed by detecting the antibodies against A/B sub- group and subgroup 3 avian leucosis virus and the p27 antigen of avian leucosis virus, ln addition,the specific sequences of exogenous ALV-A,B and J subgroup were used as primers to detect the virus in the liver samples of some p27 positive cases by PCR test. At the same time,histopathological analysis was performed. The results showed that the positive rate of ALV-A/B antibody was 5% in the 92 samples in- vestigated,the positive rate of ALV-J antibody was 36~,and the positive rate of p27 antigen in avian leucosis was 74%. The total RNA of the livers of i0 p27- positive chickens was extracted and detected by PCR test. The results showed that there was one positive case of ALV-A and ALV-B,respeetively and one positive case of ALV-J. Moreover,in these lO cases,histopathological examination revealed that there was a case of lymphocyte tumor lesions in the liver,kidney and spleen and a case of myeloid tumor le-sions in the liver. These results indicate that there are cases of infection in different virus subgroup of avian leucosis in Xinyang.
