摘  要:目的为选育替考拉宁的高产菌株。方法采用常压室温等离子体(ARTP)技术对替考拉宁产生菌Actinoplanes teichomyceticus FIM-68的孢子进行诱变,诱变处理的孢子悬液涂布在含替考拉宁致死浓度的培养基平板上培养,获得替考拉宁抗性突变株,通过摇瓶发酵对替考拉宁抗性基因突变株进行筛选。结果获得一株遗传性状稳定的替考拉宁高产菌Actinoplanes teichomyceticus FIM-68-66菌株,其产替考拉宁能力提高了2倍。结论这是首次报道采用常压室温等离子体诱变技术有效提高替考游动放线菌产替考拉宁能力,所获得的替考拉宁高产菌株可望用于替考拉宁的工业化生产。Objective To screen high-yield teicoplanin producing strains. Methods After being treated by the Atmospheric and Room Temperature Plasma (ARTP) irradiation, spore suspension of teicoplanin producing strain Actinoplanes teichomyceticus FIM-68 was spreaded on the regenerating plate containing lethal concentration of teicoplanin. Teicoplanin-resistant mutants were picked up and screened by the shake flask fermentation. Results A high-yield mutant strain Actinoplanes teichomyceticus FIM-68-66 with genetic stability was obtained. The teicoplanin production capacity of the mutant is 2 times higher than the original strain. Conclusion It was the first report that ARTP mutation was adopted to improve the capacity of teicoplanin production of Actinoplanes teichomyceticus effectively. The high-yield teicoplanin producing strain obtained was expected to be used in the industrial production of teicoplanin.
