摘 要:(1)目的探讨低浓度二甲基亚枫(DMSO)对RANKL诱导的破骨细胞前体RAW264.7细胞破骨分化的影响。(2)方法将细胞接种在24孔板上,采用0、0.001%、0.005%、0.01%、0.05%、0.1%、0.15%、0.2%等八种低浓度的DMSO作用于RANKL诱导的RAW264.7,细胞4天后,各培养孔随机采集照片20张,用显微镜每孔随机收集20张照片,使用Image-Pro Plus6.0软件对每张图进行测量,记录每张照片中RAW264.7细胞的细胞总数及总面积。(3)结果较RANKL组相比,DMSO浓度为0.005%和0.01%时,RAW264.7细胞形成破骨细胞的表面积均显著增大,差异有统计学意义(P<0.01),且0.005%和0.01%两组间差异无统计学意义(P>0.05);而DMSO浓度进一步提高为0.1%,0.15%,0.2%时,破骨细胞表面积随浓度升高而减小;DMSO浓度为0.001%,0.05%,0.1%时,破骨细胞表面积与未加DMSO组差异无统计学意义(P>0.05)。(4)结论 DMSO浓度为0.005%和0.01%时促进RAW264.7细胞的破骨分化,而随着DMSO浓度的进一步提高,DMSO对RAW264.7破骨分化的促进作用消失,甚至表现为抑制破骨分化。Objective To explore the effect of low concentration of DMSO on osteoclast differentiation induced by RANKL in osteoclast precursor RAW264.7 cells.Methods The RAW264.7 cells were seeded with 0,0.001%,0.005%,0.01%,0.05%,0.1%,0.15%,and 0.2% eight kinds of low concentration of DMSO on RANKL induced in 24 well plates.After 4 days,each well random collected 20 photographs by microscopy.The photographs were measured by Image-Pro Plus 6.0 software and recorded the total number and the total area of RAW264.7 cells in each of photos.Results Compared with RANKL group,after stimulated by 0.005%and 0.01%concentrations of DMSO,the total osteoclasts surface area was significantly larger(P <0.01),no significant difference between the two groups(P >0.05).However,the total surface area of osteoclasts formed by RAW264.7 cells decreased significantly after stimulated by 0.1%,0.15 and 0.2% concentrations of DMSO and the surface area of osteoclasts decreased as the concentration increased.Compared with RANKL only group,the total osteoclasts surface areahad no significant difference when the concentration of DMSO was 0.001%,0.05% and 0.1%(P >0.05).Conclusion The 0.005% and 0.01% concentrations of DMSO promoted the osteoclast differentiation of RAW264.7 cells,and with the further increased of DMSO concentration,the promoting effect of DMSO on the osteoclast differentiation of RAW264.7 disappeared,and even inhibited the osteoclast differentiation.
