摘  要:目的本研究探讨NOS2基因单核苷酸多态性(single nucleotide polymorphism,SNP)与单纯尘螨过敏性鼻炎(house dust mite-sensitive allergic rhinitis,HDM-sensitive AR)发生的相关性。方法研究募集了HDM-sensitive AR患者385例和健康对照受试者424例。采集外周静脉血,提取基因组DNA。Sequenom i PLEX Mass ARRAY质谱法对NOS2基因上的6个标签SNPs(rs2297516,rs7406657,rs3794764,rs2872753,rs2779249,rs3794766)进行基因分型。结果等位基因分析显示6个SNPs的等位基因频率在HDM-sensitive AR组与正常对照组间差异无统计学意义。基因型分析显示rs3794764的(AA+AG)基因型分布在HDM-sensitive AR患者组和正常对照组间差异存在统计学意义(P=0.047),相对GG基因型是HDM-sensitive AR的危险因素(OR=1.413,95%CI:1.005~1.987)。多元线性回归分析显示在健康对照人群中rs3794766基因型与血清总Ig E浓度具有显著相关性(P=0.019,β=-0.131,95%CI:-0.229^-0.021)。结论 NOS2基因可能是中国汉族人群HDM-sensitive AR的易感基因。Objective To investigate the association between single nucleotide polymorphisms( SNP) in NOS2 gene and house dust mite-sensitive allergic rhinitis( HDM-sensitive AR). Methods A total of 385 HDM-sensitive AR patients and 424 control subjects were enrolled and genomic DNA was extracted from peripheral blood leucocytes of each participant. A total of 6 reprehensive marker SNPs in NOS2 were genotyped using Sequenom i PLEX Mass ARRAY platform. Results The allele frequency analysis did not show any significant associations between 6 SNPs in NOS2 and HDM-sensitive AR. The genotype analysis showed the significant difference in distribution of rs3794764_( AA + AG) between HDM-sensitive AR patients and controls( P = 0. 047). The rs3794764_( AA + AG vs GG) presented a risk among HDM-sensitive AR patients( OR = 1. 413,95% CI: 1. 005-1. 987). Multiple linear regression analysis indicated that only rs3794766 in NOS2 gene was significantly associated with total serum Ig E levels in control group( P = 0. 019,β =-0. 131,95% CI:-0. 229--0. 021. Conclusion NOS2 genes variabilities might be associated with HDM-sensitive AR susceptibility in Han Chinese.
