ERK Pathway Involved in the Parthenogenetic Activation of Unfertilized Silkworm Eggs

摘  要:U0126是激酶MEK1/2的特异性抑制剂,可阻止下游MAPK激酶磷酸化,导致减数分裂阻滞机构失活,卵细胞恢复分裂。本实验以抑制剂U0126注射羽化前2天的雌蛹,羽化后孤雌生殖诱导未受精卵,并调查蚕卵的发育情况。结果表明:对孤雌生殖发生率不高的Nastari品系,U0126可极显著提高孤雌生殖发生率;对孤雌生殖发生率高的品系无14,U0126可部分提高孤雌生殖发生率,暗示ERK途径参与蚕卵孤雌激活。U0126 is the highly selective inhibitor of kinase MEK 112, which catalyze the phosphorylation of MAPK. Dephosphoration of MAPK led to the recovery of oocyte from meiosis arrest. In this study, inhibitor U0126 was injected into the female pupae 2 days before molt. Unfertilized eggs were harvested and parthenogenetic activated. The results showed that U0126 can improve the pigmented eggs in Nastari strain (p〈0.01) and wu 14 strain, implying that the ERK pathway were involved in the silkworm parthenogenetic activation.
