摘 要:目的研究有机磷暴露接触对内质网的影响。方法大鼠持续60天氧乐果经口染毒,检测骨骼肌中的PERK,BiP,p-eIF2α,eIF2α,IRE1α的表达水平。结果染毒组的BiP,PERK和IRE1α的表达水平均高于Control组(P<0.05)。染毒组中BiP,PERK和IRE1α的表达水平中剂量组(MD)和高剂量组(HD)均高于低剂量组(LD)(P<0.05);与MD组相比,HD组中BiP,PERK和IRE1α的表达水平较高(P<0.05)。染毒组p-eIF2α/eIF2α水平均高于Control组(P<0.05)。与LD组和MD组相比,HD组中p-eIF2α/eIF2α的水平较高(P<0.05);LD组与MD组的peIF2α/eIF2α水平的差异不具有统计学意义(P>0.05)。结论氧乐果暴露接触能够引起大鼠体内产生内质网应激。Objective To explore the effects of organophosphate exposure on endoplasmic reticulum. Methods The rats were exposed to omethoate for 60 days through gastric injection, and the expression levels of PERK,BiP, p-eIF2α, elF2α,IREIα in muscles oi rats from different groups were determined and compared. Results The expression of BiP, PERK and IREla in muscles of rats exposed to omethoate was significantly higher than that of normal rats (P〈0.05). Compared with LD group,the expression of BiP,PERK and IREla in muscles of rats from MD and HD groups was sig- nificantly increased. The expression of BiP,PERK and IREI~ from HD group was significantly higher than that of MD group (P〈0.05). The expression of p-eIF2α/eIF2α in muscles of rats exposed to omethoate was significantly higher than that of control group (P〈0.05) and it was significantly higher of HD group than that of MD and LD groups (P〈0.05). There was no significant difference in p-eIF2a/eIF2a between MD and LD group. Conclusion Exposure to omethoate can cause endoplasmie reticulum stress.
