摘  要:目的:观察人骨髓间充质干细胞(MSCs)对食管鳞癌细胞Eca-109和TE-1上皮间质转化的影响。方法:收集MSCs培养上清,分别用MSCs上清及无血清无双抗培养液作为对照诱导培养食管鳞癌细胞Eca-109和TE-1,观察肿瘤细胞形态学改变;利用qPCR、western blotting法检测Eca-109和TE-1细胞在诱导培养过程中上皮间质转化相关指标E-钙黏素(E-cadherin)、N-钙黏素(N-cadherin)和波形蛋白(Vimentin)的mRNA及蛋白水平的表达变化;利用Transwell实验检测MSCs培养上清对Eca-109和TE-1细胞迁移能力的影响。结果:MSCs上清能诱导Eca-109、TE-1肿瘤细胞形态从上皮细胞形态向间质细胞形态转化。与对照组相比,实验组上皮间质转化相关指标E-cadherin mRNA表达降低,N-cadherin和Vimentin mRNA表达升高;实验组TE-1细胞E-cadherin蛋白表达降低,Vimentin表达升高。Transwell实验显示实验组肿瘤细胞迁移能力明显增强。结论:MSCs能诱导食管鳞癌细胞Eca-109和TE-1发生上皮间质转化,增强食管癌细胞的迁移能力。Objective: To explore the effect of human bone marrow mesenchymal stem cells(MSCs)on epithelial mesenchymal transition in esophageal squamous cell carcinoma(ESCC)Eca-109 and TE-1 cells. Methods:The Eca-109 and TE-1 cells were cultured with MSCs supernatant(MSCs conditioned medium,MSCs-CM).The cellular morphology was observed under a microscope. The mRNA and protein expressions of E-cadherin,N-cadherin and Vimentin were determined by qPCR and western blotting,respectively. Transwell assay was used to detect the migration ability of cells. Results: Compared with the control group,the cellular morphology of experimental group showed mesenchymal-like changes. In response to MSCs-CM,there was decreased E-cadherin but increased N-cadherin and Vimentin at mRNA and protein levels,compared with the control group(P〈0. 05). The migration ability of the experimental group was also elevated. Conclusion: MSCs could promote epithelial mesenchymal transition in ESCC Eca-109 and TE-1 cells,and enhance the migration ability of Eca-109 and TE-1 cells.
