丹皮酚对单纯疱疹病毒性脑炎小鼠TNF-α、IL-6表达的影响

摘　　要：目的:观察丹皮酚对单纯疱疹病毒性脑炎(HSE)小鼠肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)表达的影响。方法:采用颅内注射法复制HSE动物模型,将BABL/c小鼠随机分为对照组和HSV-1感染组,分别于颅内注入20μL DMEM培养基和HSV-1病毒液(LD50,10^(-4)/20μL),并于造模后第1天起开始给予等体积丹皮酚(30 mg/kg·d^(-1))或生理盐水灌胃;经不同处理后,分为DMEM+盐水组、DMEM+丹皮酚组、HSV-1+盐水组、HSV-1+丹皮酚组,每组10只小鼠,观察各组小鼠发病情况,并在第5天断头取右侧颞叶脑组织,用酶联免疫吸附法(ELISA)测定TNF-α、IL-6的含量。结果:与对照组比较,HSV-1感染组小鼠脑组织TNF-α、IL-6含量表达明显升高(P<0.05);丹皮酚干预后TNF-α和IL-6表达均明显降低(P<0.05)。结论:丹皮酚对HSE具有一定的抗炎作用。Objective： To observe the effect of paeonol on the expression of tumor necrosis factor （TNF）-α and interleukin （IL）-6 in herpes simplex encephalitis （HSE） mice. Methods： BABL/c mice were randomly divided into two groups： the control group in which the mice were intracranially injected with 20 μL DMEM, and the HSV-1 group in which the mice were intracranially injected with HSV-1 virus solution （LD50, 10-4/20 μL）. Subsequently, the mice in the control group were sub-divided into DMEM＋normal saline （NS） group and DMEM＋paeonol group, and those in the HSV-1 group were sub-divided into HSV-1＋NS group and HSV-1＋ paeonol group （n= 10 per group）. The levels of TNF-α and IL-6 in right temporal brain tissues were measured by enzyme-linked immunosorbent assay （ELISA）. Results： The expression levels of TNF-α and IL-6 in HSV-1 group were significantly higher than those in the control group （P〈 0. 05）. However, the levels of TNF-α and IL-6 were markedly decreased after paeonol intervention （P〈0. 05）. Conclusion： Paeonol had anti-inflammatory effect on HSE in mice.
