摘  要:目的建立同时测定三七及其制剂中8种不同极性的皂苷质量分数的HPLC法。方法色谱柱:Merck Chromolith~ Performance RP-18e(100 mm×4.6 mm,macropore 2μm,mesopore 13 nm);流动相:流动相A为乙腈,流动相B为水,梯度洗脱(0~20 min,18%A;20~30 min,18%~30%A;30~45 min,30%A;45~48 min,30%~38%A;48~57 min,38%~18%A);流速:1.0 mL/min;进样量:10μL:检测波长:203 nm;柱温:30℃。结果三七皂苷R_1和人参皂苷Rg_1、Re、Ro、Rb_1、Rb_3、Rd、20(S)-F_18种皂苷分离度良好,线性范围分别为0.06~21.50μg(r=0.999 6)、0.09~36.08μg(r=0.999 6)、0.05~20.56μg(r=0.999 6)、0.02~4.08μg(r=0.999 6)、0.04~16.60μg(r=0.999 5)、0.03~8.90μg(r=0.999 4)、0.02~6.22μg(r=0.999 8)、0.03~12.58μg(r=0.999 6),三七皂苷R_1和人参皂苷Rg_1、Re、Ro、Rb_1、Rb_3、Rd、20(S)-F_1的检测限分别为16.2、27.1、15.4、3.06、12.4、6.7、4.6、9.4 ng。结论所建方法简便、准确、分离效果好、无干扰,可同时分离三七及其制剂中三七皂苷R_1和人参皂苷Rg_1、Re、Ro、Rb_1、Rd、Rb_3、20(S)-F_18种皂苷,其中5种皂苷成分可以定量。Objective To establish a new approach using HPLC with a monolithic column for determination of 8 saponins in Notoginseng and its preparation simultaneously. Methods HPLC was used with Merck Chromolith~ Performance RP-18 e( 100 mm × 4. 6 mm, macropore 2 μm, mesopore 13 nm). The mobile phase A and B were acetonitrile and water with gradient elution( 0 ~ 20 min,18% A; 20 ~ 30 min,18% ~30%A; 30 ~ 45 min,30%A; 45 ~ 48 min,30% ~ 38%A; 48 ~ 57 min,38% ~ 18%A),and the flow rate was 1.0 mL/min. The injection volume was 10 μL,the detection wavelength was set at 203 nm,and the column temperature was at 30 ℃. Results Ginsenoside Rg_1,Re,Ro,Rb_1,Rb_3,Rd,20(S)-F_1 and notoginsenoside R_1 in Panax Notoginseng and its preparation were separated at baseline with good linearity within the range of 0.06 ~ 21.50 μg( r = 0. 999 6),0. 09 ~ 36. 08 μg( r = 0. 999 6),0. 05 ~ 20. 56 μg( r = 0.999 6),0. 02 ~4.08 μg( r = 0.999 6),0. 04 ~ 16. 60 μg( r = 0. 999 5),0. 03 ~ 8. 90 μg( r = 0. 999 4),0.02 ~ 6. 22 μg( r =0.999 8),and 0.03 ~ 12.58 μg( r = 0.999 6),with a detection limit of 16.2,27.1,15.4,3.06,12.4,6.7,4.6,and 9.4 ng,respectively. Conclusion The method is simple,accurate and well separated,which could be used for determination of 5 saponins in Notoginseng and its preparation.
