摘  要:目的:采用代谢组学方法研究肝郁证哮喘模型的尿液中代谢物含量的变化。方法:采用慢性束缚加卵蛋白致敏法建立肝郁证哮喘模型。收集大鼠尿液并采用液相色谱质谱联用仪分析,将数据导入SIMCA-P13.0软件进行主成分分析和偏最小二乘判别分析,观察各组大鼠尿液的代谢组分变化。结果:鉴定出14种能够反应肝郁证哮喘模型代谢组学表征的生物标志物,乳酸、柠檬酸等7种代谢物含量较正常组降低,乙酰乙酸、肌酸酐等7种代谢物含量较正常组升高。结论:通过病证结合、以方测证的方式验证了肝郁证哮喘模型的可靠性,并鉴定出能够反映模型代谢特征的生物标志物,为中医治疗肝郁证哮喘的作用机理、作用靶点提供了依据。Objective:To study the change of urine metabolomics for asthma of liver-depression syndrome modle based on metabolomics method. Methods: Asthma of liver-depression syndrome modle was established by inhaling atomized ovalbumin and chronic restraint. The collected urine was analyzed by LC- MS/MS, and the acquired data were analyzed using principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) by SIMCA-PI3.0 software, observed the concentration changes of metabolics in each group. Results: 14 characteristic biomarkers of asthma of liver-depression syndrome modle were identified, the content of lactic acid, citric acid and other 7 kinds of metabolites were lower than those of the normal group, the content of acetoacetic acid, creatinine and other 7 kinds of metabolites were higher than those of the normal group. Conclusion : The asthma of liver-depression syndrome modle' s reliability was verified by taking a square to test certificate and combination of disease and syndrome. The identified biomarkers can reflect the metabolic characteristics of asthma of liver-depression syndrome modle, of which reveal the target and mechanism for treatment of asthma of liver-depression syndrome by Traditional Chinese Medicine.
