摘  要:目的探讨补肾健脾活血方对高糖环境下MC3T3-E1细胞的成骨分化和凋亡的影响。方法以MC3T3-E1细胞为研究对象,制备补肾健脾活血方含药血清,实验分为空白对照组、高糖组、中药低剂量组、中药中剂量组、中药高剂量组、二甲双胍组。采用流式细胞术检测MC3T3-E1成骨细胞凋亡,蛋白质免疫印记技术(Western-blot)检测来观察成骨分化相关蛋白的表达。结果与高糖组相比,中药组对高糖(25mmol/L)条件下MC3T3-E1细胞能够一定程度上抑制成骨细胞凋亡,以中药中剂量组效果更为显著(P<0.01);高糖条件下的中药组能够促进成骨分化相关蛋白BMP2和Runx2的表达,以中药高剂量组效果较为显著(P<0.01)。结论补肾健脾活血方能够促进高糖环境下MC3T3-E1成骨细胞分化和抑制凋亡的作用。Objective Discussion on the Bushen Jianpi Huoxue Formula on high glucose environment MC3T3 -E1 cell the of differentiation and apoptosis. Methods With MC3T3 -E1 cells as the research object,the experiment was divided into blank control group, the high glucose group, low dose group in traditional Chinese medicine (TCM), middle dose group in TCM, high dose group in TCM, the mefformin group. Using flow cytometry detection of MC3T3 E1 osteoblast apoptosis,immune protein imprinting technology (Western blot) test to observe osteogenesis differentiation related protein expression. Results Compared with the high glucose group, Chinese medicine group to a certain extent can inhibit osteoblast apoptosis, middle dose group in TCM effect more significant (P 〈 0.01 ). And then, Chinese medicine group can promote osteogenesis differentiation related protein expression of BMP2 and Runx2, high dose group in TCM effect more significant (P 〈0.01 ). Conclusion The Bushen Jianpi Huoxue Formula can promote glucose environment on MC3T3 -Elcell differentiation of functions and inhibit osteoblast apoptosis.
