摘  要:目的探讨西红花苷干预对急性低氧72小时大鼠脑海马FOXO3α表达的影响。方法 90只健康雄性SD大鼠随机分为5个组:正常对照组,低氧模型组,西红花苷低(25mg/kg)、中(50mg/kg)、高(100 mg/kg)剂量组。采用RT-PCR技术检测脑海马FOXO3αmRNA表达,免疫组化与Western blot蛋白免疫印迹法检测脑海马FOXO3α蛋白表达。结果 RTPCR、免疫组化及Western blot结果均显示:大鼠脑海马FOXO3αmRNA及蛋白表达量在低氧模型组高于正常对照组(P<0.05),西红花苷干预各剂量组均低于低氧模型组,其中以中、高剂量组作用显著(P<0.05)。结论急性低氧刺激后大鼠海马FOXO3α表达增强,西红花苷干预对其表达有明显的抑制作用,并发挥神经保护作用。Objective To investigate the effect of Crocin in advance of the medication on the FOXO3α expression in hippocampus at acute high altitude hypoxic environment at 72h in rats. Methods 90 SD rats were randomly divided into five groups, normal control group, hypoxia model group and low - , mid - , and high -dose (25, 50, and 100 mg/kg) Crocin groups. FOXO3α mR- NA was detected by RT - PCR technology. FOXO3α protein was detected by Immuno - histochemical and Western blot technolo- gy. Results RT- PCR, Immuno- histochemical and Western blot all showed that FOXO3α mRNA/ protein in hippocampus of rats in hypoxia model group was higher than normal control group, Crocin groups were lower than that of hypoxia model group ( P 〈 0.05), and the expression levels in mid - and high - dose groups were obviously lower (P 〈 0.05). Conclusion The expres- sion of FOXO3α in hippocampus was increased under acute high altitude hypoxia conditions, Crocin pretreatment can significantly down - regulate the expression of FOXO3α and protect neron in hippocampus from high altitude hypoxia injury.
