DRD4基因rs752306多态性与蒙古族学龄儿童攻击行为的相关性研究

摘　　要：为了解DRD4基因rs752306多态性与蒙古族学龄儿童攻击行为的相关性,选取内蒙古赤峰市天山镇蒙古族实验小学302名7~12岁蒙古族儿童为调查对象,运用Achenbach儿童行为量表(CBCL)评价了儿童攻击行为得分;采集所有儿童口腔上皮细胞并提取DNA,采用iMLDR法测定了位点基因型;利用广义线性模型(GLM)检验了主效应及交互作用大小。研究结果发现:DRD4基因rs752306位点有三种基因型GG、GA、AA,携带A等位基因的蒙古族学龄儿童攻击行为得分高于其他两种基因型;蒙古族学龄儿童年龄和父亲职业对蒙古族儿童攻击行为有影响,差异具有统计学意义(P<0.05);蒙古族儿童年龄和rs752306多态性的主效应及两者的交互作用对蒙古族儿童攻击行为有显著影响(P<0.05)。因此,DRD4基因rs752306多态性与蒙古族儿童攻击行为问题可能存在一定相关性。To investigate the association between rs752306 polymorphism in the DRD4 gene and the aggressive behav- ior in the Mongolian school-age children,a total of 302 Mongolian school--age children aged 7-12 years were se- lected as respondents from Mongolian Experimental Primary School, Chifeng, Inner Mongolia. The Achenbach Child Behavior Checklist （CBCL） was used to evaluate the scores of aggressive behavior. The DNA was extracted from oral epithelial cells and the genotyping at rs752306 was performed by improved multiple ligase detection reaction. The generalized estimating linear model was used to analyze the main effect and interaction of rs752306 polymorphism and family environment on aggressive behavior in Mongolian school--age children. The results showed that there were three genotype which were the GG, GA and AA in the rs752306 of DRD4 gene. The children with AA genotype had significantly higher scores of aggressive behavior than those with GA and GG genotype. The age of children and their father＇s occupation were significantly association with the scores of aggressive behavior in Mongolian children （P〈0. 05）. There were the significant main effects of rs752306 polymorphism in the DRD4 gene and age group on aggres- sive behavior score as well as a significant interaction of rs752306 polymorphism and age group on it （P〈0.05）. In conclusion, rs752306 polymorphism in the DRD4 gene may have correlation to aggressive behavior of Mongolian school-- age children.
