摘  要:目的观察异丙肾上腺素(ISO)诱发大鼠心肌缺血时,不同病程模型大鼠心电图和心肌P2X3受体mRNA表达的对应变化。方法50只大鼠皮下注射60mg/kgISO(连续2次,间隔24h)的方法建立心肌缺血损伤组,并以病程1、2、3、4、5d再分成5组,每组10只。另选取注射等量生理盐水的大鼠50只作为对照组。运用心电图和qPCR检测方法观察同一病程大鼠心电图波形S—T段和心肌P2X3受体mRNA的表达。结果随病程进展,模型大鼠心电图波形中S—T段呈先抬高后下降趋势,心肌P2X3受体mRNA的表达亦为先上升后下降,两者显著相关,且差异具有统计学意义。结论心肌缺血损伤大鼠心肌P2X3受体mRNA的表达增加程度和心电图波形中S—T段抬高幅度明显相关,表明心肌缺血时P2X3受体敏感度显著增强,并参与了心肌缺血的病理过程。Objective To observe the corresponding changes of ECG and myocardial P2X3 receptor mR- NA expression in rats with different stages of myocardial ischemia injury induced by isoproterenol (ISO). Methods The rat model of myocardial ischemia injury was established by subcutaneous injection of 60 mg/kg ISO(2 consecutive times, each time Interval of 24 h), and the expression of ECG S-T segment and myocardial P2X3 re- ceptor mRNA was observed by ECG and QPCR detection method in each stage of myocardial isehemia injury in every 10 rats after finishing the inject ISO of the same day (Stage 1 ), the second day (Stage 2), the third day (Stage 3 ), the fourth day(Stage 4) and the fifth day(Stage 5 ). Results The ECG S-T segment of different stages of myoeardial ischemia injury and the expression of myocardial P2X3 receptor mRNA all increased at first and then deereased which showed some correspondence with significant difference. Conclusion The expression of myocardial P2X3 receptor mRNA in different stages of myocardial isehemia injury and the amplitude of the ECG S-T segment is obviously correlated. The sensitivity of P2X3 receptor is significantly enhanced during myocardial isehemia and participated in the pathological process of myocardial ischemia.
