摘 要:采用real-time PCR检测生长激素释放肽(Ghrelin)及其受体(GHS-R)和酰基化转移酶(GOAT)在羔羊下丘脑、垂体、瘤胃、皱胃底、皱胃窦、十二指肠、空肠、回肠、心、肝、脾、肾、胰腺、睾丸的mRNA表达水平。结果显示,Ghrelin、GHS-R和GOAT mRNA在羔羊各组织中均有表达,其中Ghrelin mRNA主要表达于皱胃底(P<0.01),其次是十二指肠和胰腺(P<0.05);GHS-R mRNA主要表达于垂体(P<0.01),其次是下丘脑(P<0.05);GOAT mRNA在皱胃底和睾丸的表达水平相对较高,均显著高于其他组织(P<0.05)。研究结果表明,Ghrelin系统在反刍动物组织中广泛分布,Ghrelin可能与GHS-R和GOAT共同参与协调羔羊生长调控、摄食等功能的调节作用,为进一步研究反刍动物体内Ghrelin的生物学功能奠定了基础。This study aimed to detect the expressions and distribution pattern of Ghrelin and its receptor(growth hormone secretagogue receptor,GHS-R) and Ghrelin O-acetyltransferase (GOAT) mRNA in hy-pothalamus, pituitary, rumen, abomasum, duodenum, jejunum, ileum, heart, liver, spleen, kidney, pancreas,testis in lambs. The real-time PCR results showed that Ghrelin,GHS-R and GOAT mRNA were expressedin the tissues. Ghrelin mRNA was mainly expressed in the abomasum fundus(P〈0. 01) ,followed by duo-denum and pancreas (P〈0.05). The expression levels of GOAT mRNA in the fundus of abomasum andtestis were higher than that in other tissues (P〈0.01) ,and GHS-R mRNA was significantly higher in thepituitary (P〈0.05). The present results indicate that Ghrelin system is widely distributed in ruminant tis-sues,and Ghrelin,GHS-R and GOAT may be involved in the regulation of growth and food intake in rumi-ants.
