摘 要:为探讨在有宿主细胞的情况下,反复冻融和不同保存温度对病毒滴度的影响,将收集的含ST细胞培养的猪细小病毒和含Vero细胞培养的伪狂犬病病毒经过不同次数的反复冻融,测定其病毒滴度。将收集的猪细小病毒液和伪狂犬病病毒液分别在4、22、-20、-80℃保存,间隔一段时间后测定半数组织细胞感染剂量(TCID_(50)),以了解病毒滴度。结果显示,含ST细胞的猪细小病毒和含Vero细胞的伪狂犬病病毒经过反复冻融3次之后,病毒滴度比未经冻融的病毒滴度分别提高了1.81lg和1.86lg。同一温度下,随着放置时间的增加,病毒滴度均呈下降的趋势。在一定时间内,不同保存温度下病毒滴度有明显区别,但在-20℃和-80℃条件下保存的病毒滴度下降比4℃和22℃条件下保存的慢。因此,在培养病毒的时候可以用反复冻融3次的方法来提高病毒的滴度。4℃和22℃只能作为病毒的短期储存温度,-20℃和-80℃对于病毒来说是合适的保存温度。The aim of this study was to investigate the effects of repeated freezing and thawing and differentstorage temperatures on virus titers in the presence of host cells. The ST-cell porcine parvovirus and thepseudorabies virus containing Vero cells were collected and subjected to repeated freezing and thawingtimes to determine the virus titers. The collected porcine parvovirus and pseudorabies virus solutions werestored at 4℃ ,22℃ ,--20℃ and --80℃ ,respectively,and the virus titer was determined by tissue culturehalf infection (TCID50) for a period of time. The results showed that the virus titers of ST-cell porcine par-vovirus and Veto-containing pseudorabies virus after repeated freeze-thawing were increased by 1.81 lg and1.86 lg, respectively, compared with the freeze-thawed virus titer. At the same temperature, with the in-crease of the time,the virus titer showed a downward trend. During a certain period of time, the virus titerin the virus solution was significantly different at different storage temperatures, but the virus titers storedat --20℃ and --80℃ was slower than that at 4℃ and 22℃. Therefore,in the cultivation of the virus can beused to freeze and thaw three times to improve the virus titer. 4℃ and 22℃ can only be used as a short-term storage temperature of the virus, --20 ℃ and --80 ℃ for the virus are the appropriate storage tempera-tures.
