摘 要:目的:山姜素或豆蔻明为药食同源植物草豆蔻中的两种主要活性组分,鉴定分析这两种活性组分与人血浆相互作用后的差异显示蛋白。方法:在模拟生理条件下,应用双向电泳技术进行蛋白分离,通过基质辅助激光解析-电离飞行时间质谱分析及Mascot Distiller软件进行蛋白搜库。结果:获得山姜素或豆蔻明与人血样相互作用后的10个差异显示蛋白,经质谱鉴定为8种蛋白,分别是补体B因子、1-抗胰蛋白酶、载脂蛋白E、间α胰蛋白酶抑制剂家族重链相关蛋白、人类补体成分补体3(C3c)、阿尔法1-抗蛋白酶、类型II角质亚单位蛋白、游离型人类载脂蛋白A-I。结论:这些差异蛋白与机体防御、增强免疫效应、蛋白代谢、抑制吞噬细胞活性、增加白细胞数目、保持载脂蛋白游离构象等生理过程密切相关,也从一定程度上说明了山姜素或豆蔻明的抗菌抗炎机理及可能的靶蛋白。Objective: Alpinetin and cardamonin are two main active components of Caodoukou (Alpinia Katsumadai), which was known as the homology of medicine and food. The research objects and the differentially expressed proteins obtained from the interaction between these two main active components and human plasma samples were analyzed and identified. Methods: 2-dimensional polyacryamide gel electrophoresis (2-DE) technique was used to extract and separate the proteins of different specimens. The software Image Master5.0 was used to compare and screen out the proteins differentially expressed. These proteins were determined by using Matrix Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry (MALDI-TOF MS) and the software Mascot Distiller for searching protein databases. Results: There were 10 proteins spots that were significantly differentially expressed, 8 of which were recognized and identified with MALDI-TOF MS and the software Mascot Distiller for searching protein databases. These proteins are Complement factor B, Alpha-1-antitrypsin, apolipoprotein, Inter-alpha-trypsin inhibitor family heavy chain-related protein, Human Complement Component C3c, Scrpin peptidase inhibitor, type Ⅱ keratin subunit protein and Chain A, Crystal structure of Lipid-Free Human Apolipoprotein A-I, respectively. Conclusion: The expression of differentially proteins from the interaction between alpinetin and cardamonin and human plasma samples were associated with some physiological processes such as body's defense system, immunity enbancing efiect, proteinmctalbolism, inhibiting macrophages activity, increasing the amount of leucocyte, keeping the free conformation of apolipoproteins, and so on. The above results can contribute to explain the antibacterial and anti-inflammatory mechanism of alpinetin and cardamonin, and find the possible target proteins in blood in some degree.
